摘要
目的探讨群居相互接触对脑梗死小鼠康复训练效果的影响及可能机制。方法 2月龄雄性C57BL/6小鼠96只,随机分为4组,假手术隔离饲养组16只,假手术配对饲养组16只,大脑中动脉缺血(MCAO)隔离饲养组32只,MCAO配对饲养组32只。假手术组小鼠行假手术,模型组小鼠行MCAO手术,术后根据分组分别行单只隔离和配对饲养。各组小鼠均于术后进行康复训练:每天进行1h转棒、平衡木及网屏训练。康复训练后第1、3、7、14天行动物行为学评分,测量血清C-反应蛋白(CRP)浓度;第14天处死小鼠,2,3,5-氯化三苯基四氮唑(TTC)染色法测脑梗死体积。结果在康复训练第7及14天,MCAO配对饲养组小鼠的行为学评分和血清CRP水平均低于MCAO隔离饲养组小鼠(均P<0.05),且假手术配对饲养组血清CRP水平低于假手术隔离饲养组;第14天时,MCAO配对饲养组和MCAO隔离饲养组小鼠脑梗死体积差异无统计学意义(P>0.05)。结论群居相互接触有助于增加康复训练效果,降低血清CRP水平,对脑梗死体积无影响。
Objective To explore the effects of social interaction on rehabilitation training outcomes of mice with ischemic infarction and the possible mechanism.Methods Ninety-six male C57BL/6mice of two months old were randomly divided into four groups:sham operation group with isolated breeding(n=16),sham operation group with paired breeding(n=16),middle cerebral artery occlusion(MCAO)group with isolated breeding(n=32),MCAO with paired breeding(n=32).After the operation,the mice were isolated in individual cages or paired with an animal in different groups.All the mice were given 1-h balancing,grasping,rotating and walking exercises every day.Behavioral assessments were made and the C-reactive protein(CRP)concentrations in blood serum were detected at 1,3,7,and 14 days after rehabilitation training.At 14 days,the animals were sacrificed and the infarct size determined by TTC staining.Results The behavior score and serum CRP level were much lower in MCAO group with paired breeding than in MCAO group with isolated breeding at 7and 14 days after rehabilitation training(both P〈0.05).The CRP level was also significantly decreased in sham operation group with paired breeding than that in sham operation group with isolated breeding(P〈0.05).There was no significant difference in the infarct size of mice between the paired mice with MCAO and isolated MCAO mice at 14days(P〉0.05).Conclusion Social interaction significantly improves the outcomes of rehabilitation training and reduces the serum CRP concentration in mice with ischemic infarction,but has no significant effects on the infarct size.
