人结肠上皮NCM460细胞电阻抗谱的实验研究

摘要 目的研究细胞容积比值(cell volume ratio,CVR)对人结肠上皮NCM460细胞电阻抗谱的影响。方法采用Agilent 4294A阻抗分析仪测量了不同浓度NCM460细胞的电阻抗值和相位角,经过幅-频曲线和相-频曲线以及Nyquist图的综合分析,提取NCM460细胞的特征参数,观察细胞容积比值对NCM460细胞电生理性能的影响。结果 1)细胞容积比值对阻抗幅值最大增量(ΔZmax)、相位角峰值(θp)、低频阻抗值(Re ZL)和阻抗虚部峰值(Im Zp)等均有影响,并它们与细胞容积比值之间呈线性相关关系;2)特征频率(fc)与细胞容积比值无关,它来源于细胞膜的界面极化效应,是NCM460细胞对交流电场响应的电阻抗谱标志性参数。结论电阻抗谱方法能够获取NCM460细胞电学特征,为进一步的结肠癌细胞电生理研究提供了观测指标与方法。 Objective To develop the effect of cell volume ratio(CVR) on the electrical impedance spectrosco- py of human colonic epithelial NCM460 cells. Methods The impedance magnitude and phase angle of NCM460 cells in the different concentration were measured with Agilent 4294 Aimpedance analyzer in the frequency range of 100 Hz - 100 MHz. The characteristic parameters of NCM460 cells were extracted, and the effect of cell volume ratio on the eleetrophysiological properties of NCM460 cells was observed with the ampli- tude-frequency curve, phase-frequency curve and Nyquist plots. Results 1 ) There were effects of cell volume ratio on the maximum increment of impedance magnitude ( △Zmax ) , the peak of phase angle ( θp ) , low fre- quency limit of the impedance values (ReZL) and the peak of impedance imaginary part (ImZp). The linear correlation between these parameters and the cell volume ratio existed; 2 ) The characteristic frequency (fc) was not related to the cell volume ratio. It originated from the interface polarization of the cell membrane, to be a marker of the NCM460 cells and represent a sign of the response to the AC electric field. Conclusion The electrical impedance spectroscopy method can obtain the electrical characteristics of NCM460 cells to provide observational parameters and methods for further electrophysiological studying on colon cancer cells.
