摘要
[目的]探究北京红地球葡萄不同部位内生菌的种类及多样性。[方法]对北京市葡萄主产区种植的红地球葡萄叶片、果梗、果肉、果皮各部位进行内生菌的分离、鉴定和多样性分析。[结果]红地球葡萄不同部位的内生菌数量存在差异,果梗中内生细菌数量最多,果肉中内生真菌数量最多;共分离获得内生细菌11株,16S r DNA序列鉴定结果表明果肉中内生细菌种类多样性高于果梗、果皮和叶片,芽孢杆菌属(Bacillus sp.)在各组织中均有分布;变性梯度凝胶电泳(DGGE)结果表明不同部位内生细菌菌群存在差异性。[结论]试验结果为葡萄内生微生态系统多样性的深入研究提供了理论依据。
[ Objective ] To research the diversity analysis of endophytic microorganisms of red globe grape in Beijing. [ Method]We isolated endophytie microorganisms from grape leaves, stems, pulps and peels which were collected from a vineyard in Beijing. Further identification and diversity analysis were carried out. [ Result] There were differences in the endophyte number of different parts of red globe grape. Endo- phytic bacteria were the maximum in stems ; and endophytie fungi were the maximum in pulp. A total of 11 endophytic bacteria strains were isolated. Based on' the 165 rDNA gene sequence aligment, the species diversity of endophytic bacteria in pulps were higher than that in leaves, stems and peels. Bacillus sp. was the dominant genus existed in all the tissues. DGGE profiles further proved the diversity of endophytic mi- croorganisms in different tissues of red globe grape. [ Conclusion] This research provides theoretical foundation for the in-depth research on di- versity of grape endophytic microorganisms.
