摘要
[目的]建立藏红花愈伤组织诱导方法。[方法]以藏红花愈伤组织诱导率、诱导时间、外观形态作为评价参数,研究了外植体、激素种类及配比、外源添加物椰乳和活性炭浓度对藏红花愈伤组织诱导的影响。[结果]适宜的外植体是藏红花种球,适宜的激素种类及配比是MS+KT 0.5 mg/L+NAA 2.0 mg/L,适宜的椰乳浓度是100 m L/L,活性炭浓度是3 g/L。[结论]建立的藏红花愈伤组织诱导体系稳定性好,增殖速度较快,为工厂化生产奠定基础。
To estabiish a callus:induction-method for Crocus sativus L. [ Method] The explant, hormone type and combination, addi- tives ( coconut milk and active carbon) concentration were optimized. The appearance, callus induction rate and induction time were taken as the evaluation parameters. [ Result] The proper explant was C. sativus seedball; proper hormone type and combination was MS + 0.5 mg/L KT + 2.0 mg/L NAA. The suitable concentrations of coconut milk and active carbon in culture medium were 100 mL/L and 3 g/L,respectively. [ Con- clusion] This callus induction system for C. sativus has good stability and rapid multiplication rate ,which lays foundation for the industrialized pro- duction.
