摘要
目的:观察防风通圣散对小鼠过敏性哮喘Th1/Th2失衡的影响。方法:将72只Balb/c小鼠随机分为6组,即正常组、模型组、地塞米松对照组(每日予2mg/kg灌胃)、防风通圣散水煎剂低中高剂量组(每日分别予2.44mg/kg、4.88mg/kg、9.76gmg/kg灌胃),每组各12只。鸡卵清白蛋白(OVA)先腹腔注射,再雾化吸入建立过敏性哮喘小鼠模型。收集支气管肺泡灌洗液(BALF),进行嗜酸性粒细胞计数,ELISA检测BALF内IL-4和IFN-γ含量,计算IFN-γ与IL-4的比值。结果:与正常组相比,模型组小鼠BALF内嗜酸性粒细胞明显增多(P<0.01);BALF内IL-4含量明显升高(P<0.01),IFN-γ含量明显下降(P<0.01),IFN-γ/IL-4明显减小(P<0.01)。与模型组比较,治疗后各组BALF内嗜酸性粒细胞数量减少、IFN-γ含量增加,除防风通圣散低剂量组外(P>0.05),其余各治疗组差异有统计学意义(P<0.01);各治疗组BALF内IL-4含量均明显下降,差异有统计学意义(P<0.01);各治疗组IFN-γ/IL-4均增加,防风通圣散高剂量组和阳性药物对照组差异有统计学意义(P<0.05,P<0.01),但防风通圣散低中剂量组差异无统计学意义(P>0.05);防风通圣散各治疗组间呈浓度正相关。结论:防风通圣散能够治疗过敏性哮喘,其作用机制可能是通过调节过敏性哮喘BALF内IL-4和IFN-γ含量,平衡Th1/Th2实现的。
Objective: To observe the effect of Fangfengtongsheng Powder( FFTSP) on Th1 / Th2 imbalance in allergic asthma with mice.Methods: Totally 72 Balb / c mice were randomlly divided into the normal group,the model group,the dexamethasone group,the low,medium,and high dose FFTSP groups 12 mice for each. After allergic asthma induced by intraperitoneal injection and atomization inhalation ovalbumin. Mice in the normal group were not gastrogavage,mice in the model group were given 0. 2 m L normal saline by gastrogavage,once a day for 7 days,mice in the dexamethasone group were given 0. 2 m L dexamethasone at the dose of 2mg / kg by gastrogavage,once a day for 7 days. Mice in the low,medium,and high dose FFTSP groups were given with FFTSP〈0. 2m L at the dose of 2. 44,4. 88,9. 76 g / kg by gastrogavage,once a day for 7 days. Collected bronchoalveolar lavage fluid( BALF) for eosinophilic cell counting.The concentration of IFN-γ,IL-4 in mice BALF were measured by ELISA,and in calculating the ratio of IFN-γ / IL-4. Results:Compared with the normal group,the eosinophilic granulocyte level in mice BALF were significantly increased in the model( P〈0.01); the expression of IL-4 in mice BALF significantly increased,and IFN-γ,the ratio of IFN-γ / IL-4were significantly decreased in the model group( P〈0. 01). Compared with the model group,in mice BALF,the eosinophilic granulocyte level were significantly decreased,and the concentration of IFN-γ were significantly increased,in the medium,and high dose FFTSP groups( P〈0. 01); the expression of IL-4 in mice BALF were significantly decreased in the FFTSP groups( P〈0. 01); the ratio of IFN-γ / IL-4were significantly increased in the high dose FFTSP group( P〈0. 05). There was statistical difference among the FFTSP groups( P〈0. 05). Conclusion: The action mechanism of FFTSP in treating allergic asthma might be achieved through regulate immune factors in BALF,and correct Th1 / Th2 imbalance.
