摘要
目的:探讨鸦胆子油对人肝癌Hep G2和Huh7细胞的增殖抑制作用及机制。方法:采用MTT法分析不同浓度鸦胆子油的增殖抑制作用,并用流式细胞仪进行细胞周期和细胞凋亡分析。结果:鸦胆子油能够使Hep G2细胞和Huh7细胞的存活率降低,Hep G2细胞24h和48h的IC50值分别为59.41μl/L和45.35μl/L,Huh7细胞24h和48h的IC_(50)值为273.43μl/L和103.52μl/L。细胞周期实验中细胞G_0/G_1期比例升高。细胞凋亡实验中Hep G2和Huh7细胞早期凋亡和晚期凋亡的比例均有增加,但晚期凋亡的细胞比例增加更大,分别为(61.47±7.74)%和(69.34±6.42)%。结论:鸦胆子油能够抑制Hep G2细胞和Huh7细胞的增殖,且通过阻滞细胞周期于G_0/G_1期诱导细胞凋亡。
Objective: To investigate inhibitory effect on human hepatoma cancer Hep G2 and Huh7 cells and explore the molecular mechanism of the action involved. Methods: By MTT method to analyze the proliferation inhibition of BJO,and flow cytometry analysis was used to investigate cell cycle and cell apoptosis. Results: BJO induced the reduction of survival rate of Hep G2 cells and Huh7 cells,IC_(50) of Hep G2 at 24 h and 48 h were 59. 41μl / L,45. 35μl / L and IC50 of Huh7 cells at 24 h and 48 h were 273. 43μl / L and 103. 52μl / L. Cell cycle experiment showed that G_0/ G_1 phase increased. In cell apoptosis experiment,apoptosis index of Hep G2 and Huh7 cells were increased both in early and late apoptosis,and the proportion of the late apoptosis cell percentage increased more,they were( 61. 47 ± 7. 74) %and( 69. 34 ± 6. 42) %. Conclusion: BJO inhibit cellular proliferation of Hep G2 and Huh7 cells and induced apoptotic cellular death via arresting cell cycle at the G_0/ G_1 phase.
