摘要 目的:建立用HPLC法测定人血浆中羟基红花黄色素A(HSYA)的浓度,比较肾功能不全及肾功能正常病人血浆中HSYA的浓度差异。方法:采集临床肾功能不全及肾功能正常病人静脉滴注85mg HSYA 1h后的血浆,优选HPLC法测定,色谱柱:Waters C18柱(250mm×4.6mm,5μm);流动相:乙腈-0.05%磷酸溶液=15∶85;流速:1.0ml/min;检测波长:400nm;进样量:20μl。结果:肾功能不全组病人HSYA平均血药浓度明显高于肾功能正常组病人[(8.78±1.86)vs(3.33±0.90)μg/ml,P<0.01]。结论:肾功能不全病人使用注射用红花黄色素时需要对其进行适当调整和监测。 Objective:To establish a HPLC method for concentration detection of hydroxysafflor yellow A(HSYA)in human plasma,and to compare HSYA concentration differences between the patients with renal insufficiency and those with normal renal function.Methods:Plasma samples of the patients with renal insufficiency and those with normal renal function were collected 1hour after intravenous infusion of HSYA.Water C18column(250mm×4.6mm,5μm)was used for separation.The mobile phase was acetonitrile-0.05% phosphoric acid(15∶85)at a flow rate of 1ml/min.The detection wave-length was400 nm and the injection volume was 20μl.Results:The average concentration of HSYA in the plasma of the patients with renal insufficiency was significantly higher than that of the patients with normal renal function[(8.78±1.86)vs(3.33±0.90)μg/ml,P〈0.01].Conclusion:Adjustment and monitoring were absolutely necessary when safflower yellow was used in the patients with renal insufficiency.
