复方木鸡颗粒对小鼠慢性肝损伤的影响

The Influence of Compound Muji Granules on Chronic Liver Injury in Mice

目的:观察复方木鸡颗粒对小鼠慢性肝损伤的作用,初步探讨其可能的作用机制。方法:采用高脂饲料、CCl4、D-氨基半乳糖盐酸盐、刀豆蛋白(ConA)建立4种慢性肝损伤模型,测定小鼠血清转氨酶(ALT、AST)水平、肝指数和肝组织中丙二醛(MDA)、超氧化物歧化酶(SOD)的水平,并做肝组织病理学检查。另高脂饲料所致的慢性肝损伤检测肝组织中三酰甘油(TG)含量,ConA所致的免疫性肝损伤取脾脏,称重,计算脾指数。结果:复方木鸡颗粒能显著降低4种慢性肝损伤小鼠的肝指数以及血清中AST与ALT(P<0.05或P<0.01)和肝组织中MDA水平(P<0.05或P<0.01),升高SOD水平(P<0.05或P<0.01)。高脂饲料所致慢性肝损伤小鼠肝组织中的TG含量显著降低(P<0.05或P<0.01),免疫性肝损伤中脾指数亦减小(P<0.05或P<0.01)。结论:复方木鸡颗粒对4种慢性肝损伤有不同程度的防治作用,其作用机制可能与抑制炎症因子的释放、抑制脂质过氧化反应、清除自由基等有关。

Objective： To observe the compound Muji granules on chronic liver injury in mice, and preliminary discuss the possible mechanism. Methods：Chronic hepatic injuries were established by injections of CCl4 , D-galactosamine hydrochloride, Concanavalin （ConA） and High fat diet into mice respectively. Then levels of serum transaminase （ALT, AST） and liver in-dex were derermined. the levels of liver tissue malondialdehyde （MDA）, superoxide dismutase （SOD） were measured as well. And do liver histopathological exami-nation. Another high-fat diet-induced chronic liver damage detected in liver tissue triglyeeri-de （TG） content,and ConA in duced immunological liver injury spleen, weighed to calculat-e spleen index. Results：Compound Muji particles significantly reduced liver indices of fourchronic liver injury in mice and serum AST and ALT （P〈0.05 or P〈0.01）, and decreased the level of MDA in liver tissue （P〈0.05 or P〈0.01）, and elevated levels of SOD （P〈0.05 or P〈0.01）. High-fat diet induced chronic liver injury in mice liver tissue TG content w-as significantly lower （P〈0.05 or P〈0.01） ; the immunological liver injury of spleen index also was reduced （P〈0.05 or P 〈0.05）. Conclusion：Compound Muji Granules have better c-ureeffect on four kind ehr-onic liver injuries, and its mechanism may be related to inhibit ing the release of inflammatory eytokines, inhibition of lipid peroxidation and scav enging free radicals and so on.
