摘要
目的:研究新西兰兔心肌缺血后血清中心型脂肪酸结合蛋白(heart-type fatty acid-binding protein,H-FABP)的动态变化,以及中药桂枝汤复合方剂的干预作用。方法:将24只兔随机分为正常组、模型组、药物组,每组8只。药物组用桂枝汤复合方剂(6g·kg^(-1)·d^(-1))灌胃14d,正常组和模型组给予等量生理盐水灌胃14d。制作急性心肌缺血模型(药物组和模型组),正常组只穿线不结扎。采用结扎左冠状动脉前降支的方法复制心肌缺血模型。ELISA法检测血清H-FABP的含量,彩色多普勒超声心动图观察心脏射血分数(EF)。结果:与正常组相比,模型组结扎后30min H-FABP浓度开始升高,4h达高峰,6h开始下降(P<0.05);与模型组相比,药物组结扎后各时间点H-FABP含量均降低,结扎后6h心脏射血分数增高(P<0.05)。结论:血清H-FABP含量随心肌缺血时间有动态变化,中药桂枝汤复合方剂的预防应用可以降低血清H-FABP含量,改善心功能状态。
Objective:To observe dynamic changes of serum Heart-type fatty acid-binding protein(H-FABP)in myocardial ischemia New Zealand rabbit and intervention effect of Guizhi soup.Method:Twenty-four rabbits were randomly divided into three group:nomal group,model group and drug group.The myocardial ischemia model was created by left anterior descending coronary artery ligation.ELISA was used to detect serum content of H-FABP,Color Doppler Echocardiography was performed to measure ejection fraction(EF).Result:H-FABP concentrations began to rise at 30 min reached a peak at 4h,and then gradually decreased at 6h(P〈0.05)after myocardial ischemia.Compared with the model group,H-FABP content is lower but EF is higher in drug group(P〈0.05).Conclusion:Serum H-FABP content dynamically changes after myocardial ischemia and treatment with guizhi soup can reduce serum H-FABP content and improve heart function.
