毛诃子提取物对小鼠肝损伤的保护作用 被引量:12

摘要 目的研究毛诃子提取物对四氯化碳(CCl4)和异烟肼(INH)诱导的肝损伤的保护作用。方法给药2000,4000 mg/kg,72 h内观察小鼠中毒迹象和死亡数;建立CCl4对小鼠肝损伤模型,检测毛诃子提取物对血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、肝匀浆总超氧化物歧化酶(T-SOD)的影响;建立INH对小鼠肝损伤模型,检测肝匀浆T-SOD活性、谷胱甘肽过氧化物酶(GSH-Px)活性、丙二醛(MDA)含量,进行组织病理学分析。结果给药2000mg/kg,72 h内小鼠无异常反应;毛诃子提取物明显降低了CCl4肝损伤小鼠血清ALT、AST活性,提高了肝脏T-SOD活性;该提取物明显提高了INH肝损伤血清T-SOD活性和肝脏GSH-Px活性,降低了肝脏MDA含量,减少了异烟肼对肝脏的破坏。结论毛诃子提取物毒性小,具有一定肝保护作用,它可以和异烟肼联用,减少其对肝脏的损伤。 Objective In this study,the hepatoprotective effect of the extract of Terminalia billerica( Gaertn.) Roxb. on Kunming mice was investigated. Methods Graded doses of the extract( 2000 and 4000 mg / kg) were administered to mice,animals were observed for symptoms of toxicity and mortality for 72 h; The carbon tetrachloride( CCl4) was used to induce the liver injury in mice,the levels of alanine aminotransferase( ALT) and aspartate aminotransferase( AST) in serum and the activities of total superoxide dismutase( T- SOD) in liver homogenates were examined; The isoniazide( INH) was also used to induce the liver injury in mice,the activities of T- SOD,glutathion peroxidase( GSH- Px) and the concentrations of MDA in liver homogenates were determined,and a histopathological analysis was performed. Results The results showed that animals injected and administered orally with extract did not exhibit any symptoms of toxicity and mortality when dosed at 2000 mg / kg; The extract could reduce the ALT and AST levels in serum of mice with CCl4- induced liver injury,enhance T- SOD activities in the liver tissue;And the extract could also increase T- SOD,GSH- Px activities,decrease the MDA concentration in the liver tissue with INH-induced liver injury,and finally decrease the INH damage to liver tissue. Conclusion The Terminalia billerica( Gaertn.) Roxb.extract with low toxicity has hepatoprotective effects on mice. It could be administered in combination with INH to reduce the INH- induced liver injury.
