耐高温型灰树花菌株筛选及供试灰树花亲缘关系分析 被引量:11

摘要 从40株供试菌株中筛选获得两株可在21.5~23.5℃条件下正常开片出菇的灰树花菌株Gr0001+3和Gr0017。其中Gr0001+3的子实体性状均优于Gr0017,菌株Gr0001+3在选育耐高温灰树花品种中可作为育种亲本。本试验采用RAPD和ISSR分子标记技术对供试的40株灰树花菌株进行了遗传多样性分析。其中,8个RAPD引物共扩增出83条特异性条带,在相异系数D=0.455处将40株灰树花分为9大类。9个ISSR引物共扩增出63条特异性条带,在相异系数D=0.63处将40株灰树花分为5大类。综合分析显示,亲缘关系聚类图在分子水平上显示了供试菌株之间的亲缘关系。从中可发现,21.5~23.5℃条件下可出菇的两个菌株间亲缘关系较近,这些都将为今后灰树花的遗传育种亲本的选配提供理论依据。 We screened two Grilfola frondosa strians Gr0001 +3 and Gr0017, which can fruit normally under 21.5-23.5 ℃, from forty candidate Grilfola frondos as trains. We observed that the fruitbody characters of Gr0001 +3 were better than that of Gr0017, so Grilfolafrondosastrain Gr0001 +3 would be chosen as a parent for breeding offsprings that are tolerent with high temperature. In our study, we employed RAPD and ISSR to analyze the genetic diversity of the forty Grilfola frondosa candidate strains. Eight RAPD primers resulted in eighty-three specific bands, and divided the forty Grilfolafrondosa strains into nine groups when the dissimilarity coefficient is 0.455. Nine ISSR primers resulted in sixty-three specific bands, and divided the forty candidate Grilfola frondosa strains into five groups when the dissimilarity coefficient is 0.63. The comprehensive analysis result showed that the clustering figure indicated the genetic relationship of the forty strains at molecular level. From the corresponding analysis, we can see that the two strains that could fruit at 21.5-23.5 ℃ have a close genetic relationship, which will supply theoretical foundation for parent choice in Grilfolafrondosa genetic breeding.
