制首乌对肾阳虚、肾阴虚证大鼠肝线粒体COX及mPTP开放的影响 被引量:5

摘要 目的:探讨制首乌对肾阳虚、肾阴虚证大鼠肝线粒体COX及mPTP开放的影响。方法:将90只SD大鼠随机分成9组,即空白组、肾阳虚模型对照组、肾阳虚高、中、低剂量组、肾阴虚模型对照组、肾阴虚高、中、低剂量组,每组10只。空白组予生理盐水。肾阳虚造模给予皮下注射氢化可的松,25 mg/kg,1次/d,连续14 d,肾阴虚造模在肾阳虚证大鼠开始造模后第8天,开始腹腔注射氢化可的松,50 mg/kg,1次/d,连续7 d。肾阳、阴虚造模后模型对照组予生理盐水,其余高、中、低剂量组分别给予制首乌高(24 g/kg)、中(12 g/kg)、低剂量(6 g/kg)灌胃3个月,提取肝线粒体,测定COX活力及mPTP开放。结果:肾阴虚证大鼠肝线粒体COX活力增强,PTP通透性减小。结论:制首乌肝毒性成分可能在肾阴虚证大鼠体内毒性减弱。 Objective: To explore the effect of "Zhishouwu (Radix Polygoni Multiflori)" on COX and PTP Open in syndrome of kidney Yang deficiency and kidney Yin deficiency in rats with liver mitochondrial. Methods: The 90 SD rats were randomly divided into blank group, model control group of kidney Yang deficiency, high, medium and low dose groups of kidney Yang deficiency, model control group of kidney Yin deficiency, high, medium and low dose groups of kidney Yin deficiency, 10 rats in each. The rats were given saline solution in blank group, and subcutaneous injection of hydrocortisone in model control group of kidney Yang deficiency, according to the dose of 25 mg/kg, once a day for 14 days. Kidney Yin deficiency group in kidney Yang deficiency syndrome in rats began to build after 8 days. We had implemented for 7 days, with the cehac injection of hydrocortisone at the dose of 50 mg/kg, once a day. Rats were given physiological saline in model control group after modle building; the rest rats were given the high (24 g/kg), medium (12 g/ kg), and low dose lavage (6 g/kg) of "Zhishouwu (Radix Polygoni Multiflori)" for three months, respectively in high, medium and low dose groups. The cox activity and MPTP open were determined with extraction of liver mitochondria. Results: The COX activity enhanced and inhibits PTP permeability decreased in groups of kidney Yin deficiency. Conclusion: The hepatotoxicity ingredients of "Zhishouwu (Radix Polygoni Multiflori)" may be weakened in kidney Yin deficiency syndrome rats.
