摘要
目的:研究黄芪与葛根配伍对糖尿病模型大鼠血糖、血胆固醇(cholesterol,CHO)、甘油三酯(triglycerides,TG)和胰腺组织脂联素(adiponectin,APN)和脂联素受体(adipo Rs)水平的影响,探讨黄芪与葛根配伍对糖尿病大鼠胰腺糖脂代谢的影响及其交互效应。方法:SD雄性大鼠按血糖随机分为正常组、模型组、中药对照组、黄芪组、葛根组和黄芪葛根汤组。用链脲佐菌素(streptozotocin,STZ)诱导糖尿病大鼠模型,造模后第30天检测血糖、血脂及胰腺APN和AdipoRs水平。运用单因素方差分析和析因设计方差分析方法分析两种药物的单独效应以及两药之间的交互效应。结果:与正常组比较,模型组大鼠血糖、血清CHO和TG升高,胰腺APN水平降低,差异均有统计学意义(P<0.05),两组胰腺AdipoRs水平差异无统计学意义(P>0.05);黄芪、葛根均可下调大鼠血糖、血清CHO和TG水平(P<0.05);黄芪可提高胰腺APN水平(P<0.05),葛根作用不明显。黄芪与葛根配伍仅对血清TG呈现交互作用,且合用效果优于单一用药。结论:黄芪葛根配伍可降低糖尿病大鼠血糖、CHO和TG,升高胰腺组织APN水平,但对AdipoRs无影响,两药配伍对下调CHO水平可产生协同增效作用。
Objective: To investigate the effect and interaction of compatibility of "Huangqi (Radix Astragali)" and "Gegen (Radix Puerariae)" on glucose and lipid metabolism based on the change of blood glucose, cholesterol (CHO) and triglycerides (TG) and the adiponectin (APN) and AdipoRs in pancreas of diabetic rats. Methods: According to glucose the male SD rats were randomly divided into normal group, model group, positive control group, Radix Astragali group, Radix Puerariae group and Huangqi Gegen decoction group. Diabetes rats were induced by STZ. The blood glucose and lipids and the APN and AdipoRs levels in pancreas was detected after 30 days. The effect of each drug and their interaction were analyzed with ANOVA and Factorial design analysis of variance. Results: Compared with normal group, blood glucose, cholesterol (CHO) and triglycerides (TG) rose and the adiponectin (APN) reduced in model group , with statistical difference (P〈0.05), and the AdipoRs difference showed no statistical significance (P〉0.05). The blood glucose, CHO and TG of diabetic rats descended significantly (P〈0.OS)in Radix Astragali group and Radix Puerariae group. Radix Astragali could decrease the APN n pancreas, and Radix Puerariae showed no effect. There was only interaction on glucose between them, and the combined effects better than monotherapy. Conclusion: The compatibility of "Huangqi (Radix Astragali)" and "Gegen (Radix Puerariae)" showed effect of decreasing glucose, CHO and TG and increasing APN, but no effect on the AdipoRs in pancreas of diabetic rats. By combining, there was better effect on decreasing CHO.
