明胶海绵吸附金黄色葡萄球菌法制备大鼠胫骨骨髓炎骨缺损模型 被引量:1

摘要 目的建立易复制、操作简单、试验费用低并且符合临床发病进程的大鼠骨髓炎骨缺损模型。方法明胶海绵填充Wistar大鼠胫骨缺损部位,注射标准金黄色葡萄球菌,骨蜡封闭骨缺损。于7、14天观察大鼠局部组织变化,CR及组织病理学检查。结果组织病理学检查示:骨膜增生14例;结构破坏18例;中性粒细胞聚集19例;死骨形成19例;炎性肉芽组织增生19例。术后14天CR检查,骨膜增生4例,结构破坏10例;骨干变宽9例;死骨形成14例;病理性骨折2例及骨髓炎骨缺损17例。19只大鼠术后14天软组织肿胀明显,有14例切口破溃并伴随分泌物流出,5只局部有液动感,1只大鼠术后6天死于肺部感染。结论明胶海绵吸附金黄色葡萄球菌法能成功建立Wistar大鼠胫骨骨髓炎骨缺损模型。 Objective To establish the rat osteomyelitic bone defect model of easy replication, simple operation, low experimental cost and that conform to the clinical development process. Methods Tibia deletion position was filled by gelatin sponge and then standard staphylococcus aureus were injected into the deletion position, at last deletion position was blocked by bone wax. To observe the change of local tissue and carry out CR and pathological examination at the seventh day and fourteenth day. Results The examination of morbid histology showed that 14 cases occurred periosteal proliferation, 18 cases occurred disorganization, 19 cases occurred neutrophilic granulocyte aggregation, 19 cases occurred sequestration and 19 cases occurred inflammatory granulation tissue hyperplasia. 14 days after the surgery, CR examination showed 4 cases occurred periosteum hyperplasia, 10 cases occurred disorganization, 9 cases occurred bone shaft broadening, 14 cases occurred sequestration, 2 cases occurred pathologic fracture, 17 cases occurred osteomyelitic bone defect.19 rats with obvious soft tissue swelling 14 days after the operation, wherein, 14 cases occurred incision rupture with secretion, and 5 cases had sense of fluid in certain areas. 1 rat died of pulmonary infection 6 days after the operation. Conclusion This method that Gelatin sponge adsorbed the staphylococcus aureus could establish tibial osteomyelitic bone defect model of Wistar rats.
