摘要 通过确定Na_2S_2O_4对EA.hy926细胞造成缺氧损伤的最佳浓度和时间点建立缺氧损伤模型,以只加Na_2S_2O4为对照组,添加不同浓度的苞叶雪莲(Saussurea obvallata)粗提物,从细胞形态和细胞生存率两个方面检测其抗缺氧作用,分析苞叶雪莲粗提物对Na_2S_2O4造成EA.hy926细胞缺氧损伤的保护作用。结果表明,苞叶雪莲粗提物从1.25 mg/m L浓度开始呈剂量依赖性的增加了缺氧模型组中EA.hy926细胞的相对存活率,在10 mg/m L浓度下与Na_2S_2O4模型组相比,极显著增加了细胞存活率,是缺氧模型组的6.3倍,表明苞叶雪莲具有很强的抗缺氧损伤能力。 The experimental model of hypoxia injury was established by determining the optimal concentration and time point that sodium dithionite (Na2S2O4) causing EA.hy926 cell hypoxia demaged. Treating the one only adding sodium dithionite (Na2S2O4) as the control group,add different concentrations of Saussurea obvallata crude to explore whether Saussurea obvallata crude has protection effect from EA.hy926 cell hypoxia injury caused by sodium dithionite (NaES2O4). Two parameters were used to determine the degree of EA.hy926 cell hypoxia injury,one was cell morphology and another one was cell survival rate. The results showed that:Saussurea obvallata crude increased in a dose-dependent EA.hy926 cell survival rate in the hypoxia injury model from the concentration of 1.25 mg/mL. And there was a significant increase in EA.hy926 cell survival rate compared with sodium dithionite (Na2S2O4) hypoxia model group, which was even more than 6.3 times than hypoxia model group. It suggests that Saussurea obvallata has a significant anti-oxidant capability.
