miRNA-31基因谱与宣威女性肺癌的相关性研究 被引量:2

摘要 目的探索miRNA参与宣威女性肺癌高发的机制。方法外科手术切除的宣威女性原发性非小细胞肺癌标本30例、宣威男性原发性非小细胞癌标本15例、其它地区女性原发性非小细胞癌7例,每例取材包括肺癌病灶及距病灶5cm以上的相应癌周正常肺组织各1份,提取组织标本的RNA和miRNA,采用高通量miRNA表达芯片技术建立宣威女性肺癌相关miRNA-31基因谱。结果肿瘤组织中宣威女性与男性肺癌患者肿瘤中miR-31表达量5P(P=0.530)和3P(P=0.650)没有统计学差异;宣威女性与其他地区女性肺癌患者肿瘤中miR-31表达量5P(P=0.250)和3P(P=0.103)没有统计学差异。宣威女性与男性相比,正常人体肺组织中miR-31中的5P(P=0.750)和3P(P=0.890)没有统计学差异;宣威女性和其他地区女性相比,正常肺组织的miR-31中的5P(P=0.202)表达量没有统计学差异,但是3P(P=0.039)表达量有统计学差异。宣威地区及其它地区肺癌肿瘤组织中miR-31的5P和3P表达量都远远高于正常组织中的表达量(P均小于0.001)。结论肺癌组织中miRNA31基因突变与性别无关,与地区无关。所有地区的肺癌肿瘤组织中miR-31的5P和3P表达量都远远高于正常肺组织中的表达量,提示肺癌组织存在miRNA31基因突变。 Objective To explore mechanisms of miRNA-31 involved in the high incidence of female lung cancer in Xuanwei. Methods The contents of miRNA-31 were tested by real-time RT-PCR,and the contents of miRNA-31 were tested in lung carcinoma tissues and normal lung tissues in 30 cases female patients,15 cases male patients in Xuanwei and 7 cases female patients in other areas of primary non-small cell lung cancer. The contents of miRNA-31 were compared between the different sex patients and the patients from different areas. Results The contents of miRNA-31 5P(P=0.530)or 3P(P=0.650)had no statistically significant difference in lung carcinoma tissues between the male and female patients in Xuanwei. The contents of miRNA-31 5P(P=0.250)or 3P(P=0.103)had no statistically significant difference in lung carcinoma tissues between the female patients in Xuanwei and other areas.The contents of miRNA-31 5P(P=0.750) or 3P(P=0.890) had no statistically significant difference in normal lung tissues between the male and female patients in Xuanwei.The contents of miRNA-31 5P(P=0.202)had no significant difference and the contents of miRNA-31 3P(P=0.039)had significant difference in normal lung tissues between in the female patients in Xuanwei and other areas.The contents of miRNA-31 5P or 3P in lung carcinoma tissues were much higher than the contents of miRNA-31 5Por 3P in normal lung tissues for the patients in Xuanwei and other areas(P〈0.001).Conclusion The miRNA-31 gene mutation in the lung carcinoma tissues is unrelated to the sex or area.The contents of miRNA-31 5P or 3P in lung carcinoma tissues are much higher than the contents of miRNA-31 5Por 3P in normal lung tissues,which suggest the miRNA-31 gene mutation exists in the lung carcinoma tissues.
