摘要
[目的]观察双醋瑞因对大白兔黄肥厚黄韧带中IL-1表达及天狼猩红检测结果的影响。[方法]将黄韧带肥厚造模成功动物分为两组,A组为对照组,B组为药物组,A组以蒸馏水灌胃,B组以双醋瑞因灌胃,剂量为3.8mg/kg,连续干预3个月。两组于第4、8周和第12周对局部黄韧带进行ELISA方法检测IL-1,病理切片天狼星红染色,在偏振光显微镜下观察胶原纤维的变化。并且在Qwin V3图像分析软件对胶原面积进行统计。[结果]在实验前期,血清IL-1含量逐渐升高,在第8周最高,之后逐渐下降。药物组血清IL-1含量在第8周和第12周与对照组差异有统计学意义。偏振光观察天狼猩红染色显示,正常黄韧带构成以弹性纤维为主,弹性纤维和胶原纤维比例约为2∶1,胶原纤维以Ⅲ型胶原为主,存在少量的Ⅰ型胶原。在对照组,胶原纤维比例逐渐增加,在第4周,Ⅰ型胶原与Ⅲ型胶原相当,在第8周,Ⅰ型胶原比例明显增加,Ⅲ型胶原逐渐减少,在第12周Ⅲ型胶原大量增生,Ⅰ型胶原比例明显减少。治疗组,第4周,Ⅰ型胶原与Ⅲ型胶原比较同正常组黄韧带,在第8周和第12周,Ⅰ型胶原比例明显增加,尤其是第12周,但是较对照组,增加明显得到抑制。[结论]双醋瑞因能够明显抑制肥厚黄韧带中炎性因子IL-1的表达,抑制胶原纤维增生,从而改善肥厚黄韧带中的组织结构,使之恢复至正常状态或接近正常状态。
[Objective] To observe the effect of diacerein on IL- 1 expression and Sirius red detection results of rabbits hypertrophy of ligamentum flavum ligamentum flavum. [Methods] The animals with already made model of hypertrophy of ligamentum flavum were divided into two groups. Group A was control group,animals received distilled water gavage and group B,as the drug group,3. 8 mg / kg diacerein intragastric was given for intervention for 3 months. The IL- 1 was detected by ELISA method at 4,8 and 12 weeks,and the changes of collagen fibers were observed in the light microscope. And V3 Qwin image analysis software was used for analysis of the collagen area. [Results] In the early stage of the experiment,the content of IL-1 increased gradually,and reached the highest in the eighth week,then decreased gradually. There were significant differences in serum IL- 1 levels between the two groups at twelfth and eighth weeks. Polarized light observation on Sirius red staining showed that normal ligamentum was conposed of elastic fiber,the ratio of elastic fiber and collagen fiber was about 2: 1,most of the collagen fiber was type III,with a small amount of type I collagen. In the control group,the proportion of collagen fibers increased gradually. At the fourth week,type I and type III collagen had almost the same amount,subsequently,the proportion of type I collagen was significantly increased in the eighth weeks. In the treatment group,type I collagen and type III collagen were the same compared with the control group at the fourth week,and the proportion of type I collagen was significantly increased in eighth weeks and twelfth weeks,especially in the twelfth weeks,but in the control group,it increased more significantly. [Conclusion] The diacerein can obviously suppress the expression of inflammatory cytokines IL- 1 in hypertrophy of ligamentum flavum and decrease the hyperplasia of collagen fiber,so as to improve the organizational structure of hypertrophy of ligamentum flavum and recover it to a normal or close to normal condition.
