摘要
目的探讨分别来源于单一小麦、玉米和多种谷物与豆组合的膳食纤维对Caco-2细胞胆固醇转运的影响。方法对人结肠腺癌细胞系Caco-2细胞株进行体外培养,形成单层模型后,培养液中加入胆固醇胶囊和不同浓度的膳食纤维;MTT比色法检测细胞的生长情况,高效液相色谱法测定细胞液中的胆固醇含量。结果在100μg/ml的膳食纤维干预浓度下,3种膳食纤维均显著减少了高胆固醇状态下Caco-2细胞单层模型的胆固醇转运(P<0.05);至实验结束时,多种谷物与豆组合膳食纤维对胆固醇转运的抑制作用要好于单一小麦膳食纤维和玉米膳食纤维(P<0.05)。结论三种膳食纤维,尤其是多种谷物与豆组合膳食纤维对Caco-2细胞单层模型的胆固醇转运有一定的抑制作用。
Objective To investigate the dietary fiber( DF) from wheat( W),corn( C),and cereal-soybean combination( CSC)on cholesterol transport in Caco-2 cells.Methods The human colon adenocarcinoma cell line Caco-2 was cultured in vitro. After the monolayer model was formed,cholesterol micelles and different concentrations of dietary fiber were added into the culture medium. The growth of cells was detected by MTT colorimetry. The cholesterol content in the cell was determined by HPLC.Results In the intervene of 100 mg / ml DF concentration,three kinds of DF were significantly reduced cholesterol transport in Caco-2 cell monolayer model under the high cholesterol concentration( P〈0.05). To the end of the experiment,the inhibition effect of the CSC dietary fiber on cholesterol transport was better than that of wheat DF and corn DF( P〈0.05).Conclusions Three kinds of DF,especially DF of cereal-soybean combination,could inhibit cholesterol transport in Caco-2 cell monolayer model.
