葡萄籽提取物对大鼠肠缺血再灌注损伤的保护作用 被引量:1

PROTECTIVE EFFECT OF GRAPE SEED EXTRACT ON ISCHEMIA REPERFUSION INDUCED INTESTINE INJURY IN RATS

摘要 目的研究葡萄籽提取物(grape seed extract,GSE)对大鼠小肠缺血再灌注(ischemia reperfusion,IR)损伤的保护作用。方法雄性SD大鼠80只,随机分为4组,即对照组、小肠I/R模型组和低、高剂量的GSE组。GSE组于术前3w分别连续灌胃20、40 mg/kg bw的GSE,对照组和模型组给予同体积的生理盐水,1/d。对照组在暴露动脉后不作阻断;模型组和GSE组阻断其系膜血管30 min后再灌注60 min。随后取大鼠空肠分别进行超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量检测,单细胞凝胶电泳法检测DNA损伤程度以及用免疫组化法检测Caspase-3蛋白表达情况。结果与模型组相比,补充GSE后,T-SOD、GSH-Px活性显著升高,MDA含量显著降低。GSE组小肠细胞DNA的拖尾现象明显好转,尾长、尾部DNA含量和拖尾率都显著低于模型组。GSE组Caspase-3表达明显减少。结论 GSE能有效保护小肠I/R损伤。 Objective To explore the protective effects of grape seed extract(GSE)on ischemia reperfusion(I/R) induced intestine injury in rats. Methods Eighty male SD rats were randomly divided into four groups with equal sizes: control group, I/R model group, low and high dose GSE groups. Rats in GSE groups were treated with 20, 40 mg/kg GSE by gavage once daily for 3 weeks, while rats in control and model groups were treated with same volume of saline. Mesenteric blood vessels of rats in model and GSE groups were blocked for 30 min followed by reperfusion for 60 min. For rats in control group, mesenteric blood vessels were not blocked. Then, the activities of total superoxide dismutase(T-SOD), glutathione peroxidase(GSH-Px) and content of malondialdehyde(MDA) in the jejunum were assessed. At the same time, DNA damage was evaluated by single cell gel electrophoresis and the protein expression of Caspase-3 was detected by immuno-histochemistry. Results Compared with model group, pretreatment of GSE significantly inhibited MDA generation and increased the activities of T-SOD and GSH-Px. The results of single cell gel electrophoresis showed that there were significant broom shaped tails of intestine cells in model group. Pretreatment of GSE could significantly improve DNA damage induced by I/R. Tail length, DNA content and amount of broom shaped tails were significantly decreased compared with those of model group. In addition, there was a significant decrease in the expression of Caspase-3 in GSE groups compared with that of model group. Conclusion GSE has protective effects against intestine injury induced by I/R in rats.
