摘要
茯砖茶生产过程中"金花"菌出现的时期,一直是茯砖茶生产者和研究者关注的问题,也是进行茯砖茶生产工艺改良必备的前提条件。本研究从已测序的"金花"菌基因组中分析获得29个特异性基因,对这些特异性基因进行引物设计和筛选,最后在SI-02588和SI-02863基因上获得引物OEC-6和OEC-10,2对引物表达效率高、特异性好,2对引物可用于全程监控"金花"菌动态变化。运用2对引物检测茯砖茶生产流程中"金花"菌,结果显示在茯砖茶生产过程中萎调至第一次渥堆均为阴性,第二次渥堆至发花11 d均为阳性,这表明"金花"菌最早出现的时期是第二次渥堆前,之后至茯砖茶成品都含有"金花"菌。本研究的开展说明了"金花"菌在茯砖茶中最早出现的时期,为茯砖茶生产工艺的改良提供了理论基础和指导。
The stage that "golden flower" (Eurotium c ristatum) appeared in the production of Fuzhuan tea all along been concerned problem by Fuzhuan Tea's manufacturers and researchers. At present study, we analysised the genome ofEurotium cristatum and got 29 specific genes, and the relevant primers for each genes were designed. the results showed that the primers of OEC-6 and OEC-10 for relative genes of KT355689, KT355690 could efficiently and specificly expressed, indicating these two genes and primers could be applied in the dynamic change detection of Eurotium cristatum) in the process of Fuzhuan Tea's production. And the results showed that from waning to the first wrappin are all feminine, the second wrappin to 11 d after mouldy spot are all positive. This suggests that the existence of Eurotium cristatum among the stages of the second wrapping to Fuzhuan tea forming. This research identified the earliest stage that Eurotium cristatum in production of Fuzhuan tea, and provide theoretical basis and guidance for the Fuzhuan's process improvement.
