蒙药复方肉豆蔻五味中6个内酯的含量测定 被引量:2

Determination of six lactones in Mongolian medicine compound nutmeg-5 by HPLC

摘要 目的建立蒙药复方肉豆蔻五味中santamarine、木香烃内酯、去氢木香内酯、异土木香内酯、土木香内酯、11,13-二氢异土木香内酯的含量测定方法。方法采用HPLC法,色谱柱:Elite C18色谱柱(4.6 mm×250 mm,5μm);柱温30℃;流动相:乙腈(A)-水(B);流速:0.8 m L/min;检测波长:204 nm。结果蒙药复方肉豆蔻五味中6个内酯能完全分离,质量浓度与其峰面积具有良好的线性关系,相关系数均大于0.999。Santamarine平均回收率为102.54%,RSD为1.02%(n=6);木香烃内酯平均回收率为103.35%,RSD为0.82%(n=6);去氢木香内酯平均回收率为100.38%,RSD为1.75%(n=6);异土木香内酯平均回收率为97.81%,RSD为1.59%(n=6);土木香内酯平均回收率为96.65%,RSD为1.98%(n=6);11,13-二氢异土木香内酯平均回收率为99.64%,RSD为1.68%(n=6)。结论 3批样品测定结果表明,该方法简便、快速、准确、专属性高,可用于蒙药复方肉豆蔻五味中6个内酯的含量测定。 Objective To establish a simultaneous quantitative determination method for six lactones( santamarine,costunolide,dehydrocostuslactone,alantolatone,isoalantolactone and 11,13-dihydroisoalantolantone) from nutmeg-5,a Mongolian medicine compound. Methods HPLC analysis was performed by an Elite C18column( 4. 6 mm × 250 mm,5 μm) at temperature 30℃ with gradient elution( acetonitrile and aqueous solution) at a flow rate of 0. 8 m L/min 204 nm. Results The six lactones were separated completedly by using the established HPLC method,and the regression equation of each lactone showed good linear correlation between concentration and peak area with the correlation coefficient more than 0. 999. Each average recovery rate with relative standard deviation( RSD) of six lactones was listed as the following: santamarine 102. 54%( RSD 1. 02%),costunolide 103. 35%( RSD 0. 82%),dehydrocostuslactone 100. 38%( RSD 1. 75%),alantolatone 97. 81%( RSD 1. 59%),isoalantolactone96. 65%( RSD 1. 98%),and 11,13-dihydroisoalantolantone 99. 64%( RSD 1. 68%). Conclusion The HPLC method was validated and found to be specific,linear,precise,and accurate. This chromatographic system can be readily used on a routine way for determination of lactones from nutmeg-5.
