摘要
目的建立高山大黄药材的质量控制方法。方法采用薄层色谱法,以白皮杉醇-4'-O-β-D-葡萄糖苷为对照品对高山大黄药材进行薄层鉴别。按照2010版《中国药典》附录IX方法对高山大黄药材进行水分、总灰分及酸不溶灰分的测定。采用HPLC法同时测定高山大黄中白皮杉醇-4'-O-β-D-葡萄糖苷和白藜芦醇-4'-O-β-D-葡萄糖苷的含量。结果 10批高山大黄中白皮杉醇-4'-O-β-D-葡萄糖苷在所建立的色谱条件下分离效果好,斑点清晰。10批高山大黄药材的水分含量在7.46%~9.14%,总灰分含量在2.72%~8.75%,酸不溶灰分含量在0.50%~2.51%。10批高山大黄药材中白皮杉醇-4'-O-β-D-葡萄糖苷的含量在1.768~70.720 mg/L范围内呈良好的线性关系(r=0.999 8),加样回收率为101.11%(RSD=0.89%);白藜芦醇-4'-O-β-D-葡萄糖苷的含量在0.416~16.640 mg/L的范围内呈良好的线性关系(r=0.999 6),加样回收率为99.68%(RSD=2.54%)。结论高山大黄药材的薄层鉴别方法和HPLC含量测定方法简便、准确、重复性好,可作为高山大黄药材的质量控制方法。
Objective To explore the method to control the quality of Rheum nobile.,a widely used Tibetan herbal medicine. Methods The herb powder of ten batches of Rheum nobile was identified by thin layer chromatography( TLC) with piceatannol-4'-O-β-D-glucopyranoside as the reference substance,and the moisture content,total ash and acid-insoluble ash were detected in accordance with Appendix IX of Chinese Pharmacopoeia 2010 Edition. The contents of piceatannol-4'-O-β-D-glucopyranoside and resveratrol-4'-O-β-D-glucopyranoside were determined by using HPLC. Results The spots of piceatannol-4'-O-β-D-glucopyranoside of ten batches of Rheum nobile were clear and well separated. The range of moisture content was from 7. 46% to 9. 14%,and that of total ash was 2. 72% to 8. 75%,and that of acid-insoluble ash 0. 50% to 2. 51%. The content of piceatannol-4'-O-β-D-glucopyranoside was showed a good linear relationship at the range of 1. 768 to 70. 720 mg/L( r = 0. 999 8) and the recovery rate was101. 11%( RSD = 0. 89%),by parallel,that of resveratrol-4'-O-β-D-glucopyranoside was showed a good linear relationship at the range from 0. 416 to 16. 640 mg/L( r = 0. 999 6) and the recovery rate was99. 68%( RSD = 2. 54%). Conclusion The methods of TLC and HPLC are simple,accurate and repeatable,which could be the option of the quality control of Rheum nobile.
