尼美舒利对肝癌细胞HepG2增殖及bcl-2和Bax表达的影响

摘要 目的研究尼美舒利对肝癌细胞增殖的影响及作用机制。方法通过MTT法检测不同浓度尼美舒利(50μg/ml、100μg/ml、150μg/ml)对肝癌细胞Hep G2 72 h增殖的影响,并通过western-blot方法检测Bax及bcl-2表达变化。结果尼美舒利可抑制肝癌细胞Hep G2增殖,并且抑制率与作用浓度呈现剂量依赖关系,Bax的表达随着尼美舒利浓度的升高而逐渐升高,bcl-2则逐渐降低,二者均与尼美舒利作用浓度呈良好的线性关系。结论尼美舒利可抑制肝癌细胞Hep G2增殖,并可增强Bax的表达,抑制bcl-2表达。 Objective To investigate the effect of nimeisulide on liver cancer cell proliferation and the possible function mechanism. Methods To detect the cell viability of Hep G2 through MTT assay after treated with nimeisulide 72 h at different concentrations(50μg/ml,100μg/ml,150μg/ml), and to detect the expression of Bax and bcl-2 by western-blot method. Results Nimeisulide could inhibit the proliferation of Hep G2 liver cancer cells,and inhibition rate presented dose-response relationship with nimeisulide concentrations,the expression of Bax increased with rising of concentration of nimesulide,but bcl-2 decedent, which both presented dose-response relationship with nimeisulide concentrations. Conclusion Nimeisulide could inhibit the proliferation of Hep G2 liver cancer cells,and also could increase the expression of Bax,but decrease the expression of bcl-2.
