摘要
目的:比较不同加工方法对杜仲桃叶珊瑚苷含量的影响,为杜仲的产地加工提供科学依据。方法:采用高效液相色谱法测定桃叶珊瑚苷量,色谱条件:Diamonsil C18色谱柱(250 mm×4.6mm,5μm),流动相甲醇-水溶液(10∶90),检测波长203nm,柱温25℃,流速1.0m L/min,进样量10μL。结果:不同加工方法杜仲桃叶珊瑚苷含量有差异。同一干燥方法,未发汗杜仲桃叶珊瑚苷量高于发汗杜仲;同一性状,烘干、晒干、阴干三者间桃叶珊瑚苷量具显著差异,以烘干最高,晒干次之,阴干最低;不同性状,板片状以100℃烘干、宽丝状以60℃烘干含量最高。结论:不同加工方法对杜仲桃叶珊瑚苷含量影响显著,临床应用如以桃叶珊瑚苷的生物活性为主,结合实际生产,建议杜仲采用趁鲜切宽丝,不发汗,60℃烘干的产地加工方法。
Objective: To provide scientific basis for the processing method of Eucommia ulmoides Oliv by comparing the effect of different processing methods on the content of aueubin. Methods: Determine the content of aucubin in Eucommia ulmoides Oliv (EUO) by HPLC. A Diamonsil C18 column (4. 6 mm ×250 mm, 5 μm) was used for the analysis. The mobile phase was methanol - water ( 10 :90), the flow rate was 1. 0 ml/min, the detection wavelength was at 203nm, and the column temperature was 25℃. Results: Eu- commia ulmoides Oliv processed by different methods, the content of aucubin in them was various. Living on the same drying methods, the content of aucubin in herbs without undergoing the diaphoretic process was higher than the herbs experienced it. Herbs in the same trait, with different drying methods, there were significant difference in the content of aucubin, and the content in the drugs dried by oven was higher than the drugs dried by exposure to the sunor inthe shade. Herbs in different trait, the content of aucubin in the plate - like EUO samples dried in the oven in 80% and the wile -filamentous -like EUO samples dried in the oven in 60℃ was both higherthan all other like samples. Conclusion: There were significant difference in the content of aucubin of different samples processed by different methods. In the clinical application, if we considered the biological activity of aucubin as a focus, we suggested the EUO should be pro- cessed in wile - filamentous - like without undergoing diaphoretic process and dried in the oven in 60℃.
