摘要
为了研究黑龙江省布鲁氏菌的遗传变异情况,试验采用PCR方法对9株黑龙江布鲁氏菌分离株的Omp25基因编码区进行了扩增,获得了该基因的核苷酸序列,并推导出其编码的氨基酸序列,将获得的序列与参考菌株进行比较,分析其序列特征和蛋白结构特征。结果表明:9个分离株之间的同源性很高,其中HLJ-5株与羊种布鲁氏菌参考毒株亲缘关系较近,其他8株与牛种布鲁氏菌参考毒株亲缘关系较近。Omp25基因编码蛋白存在1个信号肽和1个跨膜区,但没有潜在的N-糖基化位点。说明黑龙江省布鲁氏菌流行菌株以牛种菌株为主,且较为保守。
To study the genetic variation of BruceUa in Heilongjiang province, the ceding regions of the Omp25 genes from the nine isolates of Brucella in Heilongjiang province were amplified using PCR assay. The nucleotide sequences of the Omp25 genes were obtained, and the amino acid sequences encoded by the genes were also deduced. And then the nucleotide sequence and deduced amino acid sequences were compared with those of reference strains, and the characteristics of the sequences and the protein structure were analyzed. The results showed that all iso- lates had high homology, thereinto, the HLJ -5 isolate had a close kinship with the reference strains of Brucella melitensis (B. melitensi), and the other eight isolates had a closer kinship with the reference strains of Brucella abortus ( B. abortus). The protein encoded by the Omp25 gene contained a Signal peptide and a transmembrane region, and there was, no potential N - glycosylation site. The results indicate that the B. abor- tus strain is dominant in the prevalent Brucella strains in Heilongjiang province, and is more conservative.
