虎斑颈槽蛇细胞色素b基因片段遗传特征分析

摘要 为了研究捕捉于吉林省白山市的1条蓝色虎斑颈槽蛇与其他同亚科游蛇的亲缘关系和其分类学地位,试验采用DNA测序技术、同源序列比较的方法提取其基因组DNA,并对其细胞色素b(Cytb)基因进行扩增并测序,然后与17种同亚科游蛇的细胞色素b基因进行同源性比较。结果表明:虎斑颈槽蛇基因突变位点未饱和,仍有变异趋势;虎斑颈槽蛇J-01变异与地理环境有关。说明虎斑颈槽蛇J-01为变异虎斑颈槽蛇,其是否可以上升为新的虎斑颈槽蛇亚种仍有待下一步研究。 To study the phylogenetic relationships between a blue Rhabdophis tigrinus that captured in Baishan city of Jilin and other snakes in the same subfamily, and its taxonomic status ,the DNA sequencing technology and the method of homologous sequence comparison were used in the test. A gonomic DNA of Rhabdophis tigrinus was extracted, and its cytochrome b (Cytb) gone was amplified and sequenced. And then the homology comparison of the Cytb gone was carried out between Rhabdophis tigrinus with 17 species of colubrids in the same subfamily. The re- suits showed that the gone mutation sites of Rhabdophis tigrinus were not saturated, and there was still a trend of variation: The variation of Rhabdophis tigrinus J-01.was related to the geographic environment. The results indicate that the Rhabdophis tigrintts J-01 belongs to variant Rhabdophis tigrinus. Whether the variant Rhabdophis tigrinus can give rise to a new subspecies of Rhabdophis tigrinus, remains to be further study.
