肺痨康与耐异烟肼结核ahpC基因突变关系的研究 被引量:1

摘要 目的:探讨肺痨康对耐异烟肼结核ahpC基因突变的影响。方法:采用全基因重测序的方法,对药物干预后的耐异烟肼结核模型小鼠肺组织结核杆菌基因组DNA进行重测序。以标准菌株(H37RV)为参考序列,筛选出与ahpC基因相关的突变位点,进行对比分析。结果:7个碱基突变位点的ahpC基因对应的密码子和氨基酸均未发生改变。肺痨康组和肺痨康联合异烟肼组的突变位点2729621,对应的密码子Gcc→Tcc,氨基酸由丙氨酸变为丝氨酸。结论:肺痨康对ahpC基因突变无修复作用,突变位点2729621,可能是肺痨康干预过的小鼠与未使用肺痨康干预过的小鼠之间的个体差异相关的单核苷酸多态性(SNP)位点。肺痨康抗结核的作用机制,需要参考与异烟肼耐药相关的其它基因的文献资料做进一步研究讨论。 Objective: To explore the effect of Feilaokang on Isoniazid-resistant TB ahpC gene mutation. Method: Used the method of whole genome sequencing after drug intervention of isoniazid resistance tuberculosis mouse with lung tissue tuberculosis DNA analysis.In standard strains ( H37RV ) as the reference sequence, selected associated with ahpC gene mutations to make a contrastively analyzed.Result: Seven bases of ahpC gene corresponding codon and amino acid had not been changed.The feilaokang group and feilaokang combine isoniazid group 2729621 of gene mutation locus, the corresponding codon Gcc changed to Tcc, by alanine to serine.Conclusion: Feilaokang can't repair ahpC gene mutation locus 2729621 maybe is the single nucleotide polymoIphism refer to individual difference between the mouse using feilaokng intervention with the mouse not using feilaokang intervention.The mechanism of feilaokang resistance TB need refer to more gene literature associated with isoniazid resistance to make further researching and discussion.
