印刷油墨挥发气体对小鼠的肝肾功能损伤

The function damage of liver and kidney in printing ink volatilization gas to mice

摘要 目的探讨吸入印刷油墨挥发物对小鼠肝肾功能的损伤。方法 30只昆明种雄性小鼠,随机分为对照组、新闻纸油墨组和铜版纸油墨组,每组10只。采用静式染毒方法,新闻纸油墨组平均挥发浓度为152mg/m3,其中苯系物浓度为69.9mg/m3(含苯2.7mg/m3),烯烃浓度为15.7mg/m3,烷烃浓度为68.5mg/m3;铜版纸油墨平均挥发浓度为270mg/m3,其中苯系物浓度为106.9mg/m3(含苯4.05mg/m3),烯烃浓度为38.8mg/m3,烷烃浓度为124.5mg/m3;对照组不加任何物质。吸入染毒2h,1次/d,每周染毒6d,染毒30d后处死小鼠。检测小鼠血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、肌酐(Cr)、尿素氮(BUN)、超氧化物歧化酶(SOD)和丙二醛(MDA)的含量,并对小鼠进行肝肾病理分析。结果新闻纸油墨组和铜版纸油墨组小鼠肝组织中ALT及MDA含量与对照组比较均升高,SOD活力均降低,差异有统计学意义(P<0.05)。新闻纸油墨组和铜版纸油墨组肾组织中BUN含量与SOD活力高于对照组,差异均有统计学意义(P<0.05)。新闻纸油墨组肝病理可见,肝组织有轻度空泡变性,肝细胞肿胀,胞浆轻度水肿;肾病理可见肾近曲小管轻度水肿,胞质疏松,肾小球尚正常。铜版纸油墨组可见肝细胞索排列轻度紊乱,中央静脉为中心出现轻度弥漫性病理改变,肝细胞有中度空泡变性,肝细胞轻度肿胀,胞质水肿;肾组织可见肾小球轻度萎缩,肾小管上皮细胞水肿,胞浆疏松。结论反复吸入油墨挥发物对小鼠的肝肾功能有损伤,但无严重的器质性病变。 Objective We discussed the printing ink volatile inhalation injury on hepatic and renal function of mice. Methods 30 male Kunming mice were randomly divided into 3 groups, control group, news paper ink group and coated paper ink group, 10 rats in each group. Using the method of static intoxication, news paper printing ink group average volatile concentration was 152 mg/m3 , the concentration of benzene was 69.9 mg/m3 (containing benzene 2.7 mg/m3 ), olefin concentration was 15.7 mg/m3 , alkane concentration was 68.5 mg/m3; coated paper ink average volatile concentration is 270 mg/m3, the concentration of benzene was 106.9 mg/ma (containing enzene 4.05 mg/m3) Olefin concentration was 38.8 mg/m3 ), alkanes concentration was 124.5 mg/m3 ; the control group without any substance. Inhalation exposure to 2 h, once each day, 6days exposure weekly, mice were sacrificed after 30 d exposure. We detected the content of AST, ALT, Cr, BUN, SOD and MDA of serum, and analysised the pathological of liver and kidney. Results The content of ALT and MDA in news print and coated paper ink group of mice liver tissue increased compared with control group, and SOD content decreased, the difference was statistically significant(P 〈0.05). The content of BUN and SOD in news paper printing ink group and coated paper ink group of mice tissue was higher than the control group, and the difference was statistically significant (P 〈0.05). News paper printing ink group liver pathological was visible, liver tissue with mild vacuolar degeneration, hepatocyte swelling, cytoplasm mild edema; Renal pathological, renal proximal convoluted tubules mild edema, cytoplasm rarefaction, glomerulus was normal. Coated paper ink group showed mild hepatic cords arranged disorder, central vein center appeared mild diffuse pathological changes, liver cells had moderate vacuolar degeneration, mild swelling of liver cells, cytoplasmic edema; kidney glomerulus atrophy, dropsy in renal tubular epithelial ceils, cytoplasm rarefaction. Conclusions Repeated inhalation of ink volatiles on mice, liver and kidney function was damaged, but not serious pathological changes.
