摘要
目的探讨BEZ235体内外抑制乳腺癌细胞的作用。方法选用人三阴乳腺癌细胞系MDA-MB-231及三阳乳腺癌细胞系MCF-7,用MTT检测细胞增殖;用裸鼠荷瘤实验分析乳腺癌细胞系体内增殖;用Western blot法检测PI3K/Akt相关信号通路蛋白的表达。结果 BEZ235对乳腺癌细胞具有明显的增殖抑制作用(P<0.01),尤其是对三阴性乳腺MDA-MB-231细胞具有较强的体内外抑制作用(P<0.01)。结论 BEZ235可体内外抑制乳腺癌细胞,其机制可能是通过抑制PI3K/Akt信号通路蛋白的磷酸化修饰而实现的。
Objective To investigate the effect of BEZ235 on breast cancer cells. Methods Proliferation ability of human triple negative breast cancer cell line MDA-MB-231 and triple positive breast cancer cell line MCF-7 was de- tected by MTI" assay ; the xenograft model to analyze the ability of exhibited breast cancer proliferation in vivo ; West- ern blot was used to detect the PI3K/Akt signaling pathway related protein level. Results BEZ235 significantly in- hibited the proliferation of breast cancer cells (P 〈 0. 01 ). Especially BEZ235 strongly inhibited the triple negative breast cancer MDA-MB-231 cells in vivo and in vitro (P 〈 0. 01 ). Conclusions BEZ235 may be through the inhibi- tion of PI3K/Akt signaling pathway proteins phosphorylation to inhibit breast cancer ceils in vitro and in vivo.
