赤芍-黄柏药对配伍前后化学成分变化研究 被引量:4

摘要 目的:利用高效液相色谱(HPLC)法对赤芍和黄柏单煎、单煎后合并及合煎进行分析,寻找异同。方法:采用Agilent Eclipse Plus C_(18)(250 mm×4.6 mm,5μm)色谱柱进行分离,流动相采用乙腈-0.2%甲酸溶液梯度洗脱,流速1.0 mL·min^(-1),柱温30℃,进样量5μL,多波长检测。结果:定量分析了不同配伍比例(1:2,1:1,2:1)中芍药苷、小檗碱、黄柏碱的含量。与单煎液相比,合煎液中芍药苷溶出量增多,且随黄柏用量增加而增多,而黄柏碱和小檗碱溶出量均减少。其中,小檗碱明显减少,且随赤芍的用量增加而减少。归属了赤芍、黄柏药对主要色谱峰的来源,定性比较了色谱峰所代表的成分在1:1合煎液与1:1合并液中峰面积,发现合煎后赤芍中单萜苷类化合物的溶出增加,黄柏中的化合物溶出有增有减。结论:赤芍和黄柏合并液及合煎液的指纹图谱有异同,表明赤芍、黄柏药对的物质基础和赤芍、黄柏单用的物质基础有异同,为进一步研究该药对的药效物质基础提供了一些试验依据。 This study was aimed to analyze the chemical changes of Chi-Shao(CS,Radix Paeoniae Rubra)-Huang-Bai(HB,Phellodendron chinense) herb pair before and after compatibility by HPLC.Agilent Eclipse Plus C18(250 mm×4.6 mm,5μm) chromatographic column was adopted.The mobile phase was acetonitrile-0.2%formic acid solution with gradient elution.The flow rate was 1.0 mL·min^-1.The column temperature was 30℃.The injected volume was 5μL.The detection was with multiple wavelengths.The results showed contents of paeoniflorin,berberine and phellodendrine in different compatibility proportions(1:2,1:1 and 2:1)with quantitative analysis.Compared with the single decoction,the content of paeoniflorin in the compound decoction was increased.Its amount increased with the increasing of HB dose.However,the contents of phellodendrine and berberine were decreased.The content of berberine decreased obviously.It decreased along with the increasing of CS dose.Major peaks were confirmed to the herb pair.Qualitative comparison on the area of chromatographic peak represented component in 1:1 compound decoction and 1:1 merge decoction.It showed that the contents of monoterpene glycoside compounds in CS were increased,some compounds in HB increased,and other compounds in HB decreased in the compound decoction.It was concluded that there were differences on the fingerprints between CS-HB merge decoction and compound decoction.There were differences on the material basis between the CS-HB herb pair and the single use.It provided some experimental evidences for the study on pharmacodynamic substances of this herb pair.
