HPLC测定FKⅣ号颗粒中芍药苷、绿原酸、虎杖苷及丹酚酸B的含量 被引量:3

摘要 目的:建立同时检测FKⅣ号颗粒中芍药苷、绿原酸、虎杖苷及丹酚酸B的HPLC含量测定方法。方法:采用HPLC含量测定法,色谱柱为Kromasil C18(4.6 mm×250 mm,5μm);以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,芍药苷检测波长为230 nm,绿原酸、虎杖苷、丹酚酸B检测波长为306 nm;柱温30℃,进样量10μL。结果:芍药在7.539-482.470μg·mL^(-1)范围内呈良好线性关系(r=1.000),平均回收率为101.10%;绿原酸在2.003-128.166μg·mL^(-1)范围内呈良好线性关系(r=1.000),平均回收率为100.76%;虎杖苷4.503-288.200μg·mL^(-1)范围内呈良好线性关系(r=1.000),平均回收率为100.74%;丹酚酸B在4.621-295.752μg·mL^(-1)范围内呈良好线性关系,平均回收率为99.91%(r=0.999 8)。结论:该方法简便可靠,重复性好,可作为FKⅣ号颗粒中芍药苷、绿原酸、虎杖苷及丹酚酸B的含量测定方法。 This study was aimed to establish the simultaneous content determination of paeoniflorin,chlorogenic acid,polydatin and salvianolic acid B in FK Ⅳ granules by HPLC.The Kromasil C18 column(4.6 mm×250 mm,5Jim) was used as chromatographic column.The acetonitrile-0.1%phosphoric acid solution was used as mobile phase with gradient elution.The detection wavelength of paeoniflorin was 230 nm.And the detection wavelength of chlorogenic acid,polydatin and salvianolic acid B was 306 nm.The column temperature was 30℃.The sample injection volume was 10μL.The results showed that paeoniflorin had a good linear relationship in the range of7.539-482.470μg·mL^-1(r = 1.000) with the average recovery rate of 101.10%.Chlorogenic acid showed a good linear relationship in the range of 2.003-128.166μg·mL^-1(r = 1.000) with the average recovery rate of 100.76%.Polydatin showed a good linear relationship in the range of 4.503-288.200μg·mL^-1(r = 1.000) with the average recovery rate of 100.74%.Salvianolic acid B showed a good linear relationship in the range of 4.621-295.752μg·mL^-1(r=0.999 8) with the average recovery rate of 99.91%.It was concluded that the method was simple and reliable with good reproducibility.It can be used as a method for the content determination of paeoniflorin,chlorogenic acid,polydatin and salvianolic acid B in FK Ⅳ granules.
