摘要
肿瘤原代细胞三维立体培养法药敏检测技术(collagen gel droplet-embedded culture drug sensitivity test,CD-DST)是一种先进的体外化疗药物敏感性检测方法 ,相对于其它方法有着无可比拟的优势,比如细胞用量少、简便快速等。自该法建立以来,大量基于此平台的临床研究便陆续开展起来,主要针对一些常见的恶性肿瘤,如大肠癌、乳腺癌等。大量研究证实,CD-DST技术的检测结果与临床治疗效果的拟合程度很高,应用该方法模拟的化疗方案,患者都取得了较好的疗效。CD-DST对于建立肿瘤患者的个体化治疗方案有着重要的指导意义,相信随着技术的不断发展以及更广泛的应用,会在个体化治疗中得到更广泛的应用。本文综述了CD-DST技术的建立及研究进展,有望为该技术的发展提供新的思路,为肿瘤患者提供新的治疗手段。
The drug sensitivity of tumor cells is one of the critical issues to conduct individualized therapy for cancer patients. One of the therapeutic approaches is in vitro anticancer drug sensitivity test. Collagen gel droplet-embedded culture-drug sensitive test(CD-DST) is one of the best chemosensitivity tests, and is apparently superior to other methods currently available, because it requires a low number of the cells, has a short turn-around time and involves a simple and easy procedure. Since its establishment, CD-DST has been extensively applied and evaluated in clinical studies for the treatment of malignant tumors. It has been demonstrated that the chemotherapeutic protocols designed according to the results of CD-DST shows a high degree of fitting to the clinical scenario with high therapeutic efficacy and remission rate. CD-DST, as a guideline for the therapeutic decision making, is indispensable in individualized chemotherapy for cancer patients, and it may achieve more extensive clinical application with advance of new techniques. In this article, we review the establishment and advance of CD-DST, hoping to provide some basis for the future study.
