摘要
目的 EGFR19基因有19种常见突变型,临床检测分析一般很难分辨是何种突变型,探讨利用Excel软件制作EGFR19突变焦磷酸测序法参考图,提高工作效率。方法根据体细胞基因突变特点,假设EGFR19的突变型基因占20%,野生型基因占80%,将突变前后序列从2235号碱基起按顺序录入并按比例依次赋值,再利用Excel强大的图表生成和函数计算功能生成峰值模型图,经优化处理后可得到参考图。结果 EGFR19的19种参考图可根据录入的数据自动描点生成相应的参考图,实验室工作人员可灵活应用其对检测结果进行基因分型判断,进而更加合理的指导临床用药。结论应用Excel制作EGFR19的参考图具有操作简单、快捷准确和灵活性等优点,参考图有利于基因检测结果的分型。
Objective The EGFR19 somatic mutation were with 19 mutation types, but clinical genetic tests and analysis could not distinguish these mutation types. To draw the pyro - sequencing reference model of EGFR19 mutation with Excell will contribute to improving work efficiency. Methods According to the characteristics of somatic gene mutation, and assumed that the mutation gene of EGFR19 accounted for 20% and the wild gane accounted for 80% , and the gene sequences and test values were sequentially imputed in Excel from the number 2 235th base, then using the chart generation and calculation function of Excel to draw peak model diagram, and after optimization, the reference chart was obtained. Results 19 kinds of reference charts of EGFR19 could be obtained by automatically tracing point according to the input data, the laboratory staff can flexibly applied genotyping judgment with the test results, and be more reasonable to guide the clinical medication. Conclusion Using Excel to draw EGFR19 reference chart model is simple, fast, accurate and flexible, and the reference model could be in favor of the the typing of gene test results.
