宫颈上皮内瘤变及宫颈癌中人乳头状瘤病毒感染与P16基因表达的关系 被引量:9

Relationship between cervical intraepithelial neoplasia,human papillomavirus infection in cervical carcinoma and the expression of P16 gene

摘要 目的研究宫颈上皮内瘤变及宫颈癌组织中人乳头状瘤病毒(HPV)感染与抑癌基因P16表达的关系。方法采用免疫组织化学法检测35例宫颈癌、45例子宫上皮内瘤变(CIN)和30例慢性宫颈炎组织的P16基因的表达情况,同时采用PCR-反向杂交法对其宫颈细胞进行HPV分型检测。结果从慢性宫颈炎、CIN到宫颈癌,各组病例HPV的感染率和P16基因的表达率均呈递增趋势,CIN组和宫颈癌组HPV感染率和P16基因的表达率显著高于慢性宫颈炎组,差异有统计学意义(P<0.01)。在CIN与宫颈癌病例组织中,P16基因的表达率与HPV感染率呈正相关(r=0.98,P<0.05)。结论在CIN、宫颈癌发生发展过程中,HPV感染和P16基因表达均发挥重要作用,而且HPV感染是导致P16基因高表达的原因之一。 Objective To study the relationship between cervical intraepithelial neoplasia( CIN), human papillomavirus (HPV) infection in cervical carcinoma and the expression of P16 gene. Methods The expression of P16 gene was detected by immuno - histochemical method, from 35 cases of cervical carcinoma, 45 cases of CIN and 30 cases of chronic cervicitis, and HPV typing of cervical cells was detected by PCR - reverse hybridization method. Results From chronic cervicitis, CIN to cervical cancer, the infection rates of HPV and the expression of P16 gene in each group were at the increasing trend, the HPV infection rate and the expression rate of P16 gene in CIN group and cervical cancer group were significantly higher than those in chronic cervicitis group, and the differences were statistically significant(P 〈0.01 ). The expression of P16 gene was positively correlated to the HPV infection rate rate in patients with CIN and cervica carcinoma(r = 0.98, P 〈 0.05 ). Conclusion The expression of P16 gene and HPV infection may play a key role in the process of CIN and cervical carcinoma, and HPV infection may be an impor- tant factor to the high expression of P16 gene.
