摘要
目的建立反相液相色谱法,同时测定3种不同种类的化妆品中17种抗生素。方法在Synergi Hydro-RP C18(4.6 mm×250 mm,5μm)色谱柱上,以甲醇-乙腈-三乙胺磷酸溶液为流动相进行梯度洗脱,流量为1.0 ml/min,检测波长为210 nm和268 nm,柱温为30℃,外标法进行定性、定量测定。结果在10μg/ml^200μg/ml时,所得17种抗生素的回归方程均呈良好的线性关系,除氧氟沙星外,相关系数均>0.995,精密度良好,相对标准偏差(RSD)≤2.5%(n=6),最低检出浓度为0.5μg/g^21.7μg/g;除氯霉素外,低、中、高3个水平的各物质平均回收率均为68%~125%。结论该方法操作简便、快捷、准确、灵敏度高,适用于化妆品中17种抗生素的同时测定。
Objective To establish a reversed phase liquid chromatography method for the simultaneous determination of 17 antibiotics in 3 different cosmetics. Methods The analysis was performed on a Synergi Hydro- RP C18column( 4. 6 mm ×250 mm,5 μm) at a flow rate of 1. 0 ml / min and the detection wavelength of 210 nm and 268 nm with the column temperature of 30 ℃,using methanol- acetonitrile- triethylamine phosphoric acid solution as mobile phase. The external standard method was employed for quantitative analysis. Results The regression equation of the 17 compounds except ofloxacin showed a good linear relationships among 10 μg / ml- 200 μg / ml,with correlation coefficients( r) 〉0. 995. The precision was good,and the relative standard deviations( RSD) were no more than 2. 5%( n = 6). The lowest detection limits were among 0. 5 μg / g-21. 7 μg / g. Except clindamycin,the average recovery rate of each material at low,medium and high levels was all among68%- 125%. Conclusion The proposed method is simple,fast,accurate,sensitive,and is applicable to simultaneous determination of 17 kinds of antibiotics in cosmetics.
