摘要
目的观察强精片对不育大鼠睾丸生精微环境血.睾屏障(BTB)的影响及可能的机制。方法70只雄性SD大鼠随机分为空白组(BG组)20只和雷公藤多甙片(GTW)灌胃的造模组50只,灌胃3周后两组各取10只进行模型验证。其余40只造模大鼠随机分为模型组(MG组)和强精片低剂量组(QJPL组)、中剂量组(QJPM组)、高剂量组(QJPH组),灌胃4周后处死分离睾丸,检测相关指标。结果与BG组相比,MG组睾丸组织中FasL、ABP表达量明显上调(P〈0.01),而紧密连接蛋白Occludin表达降低(P〈0.01)。应用药物强精片干预后,FasL、ABP蛋白在睾丸内表达降低,其中高剂量组更为显著(P〈0.01);Occludin蛋白较MG组表达上调,差异具统计学意义(P〈0.05)。结论通过上调睾丸组织内FasL、ABP蛋白的表达,改变支持细胞功能,同时抑制Occludin蛋白的表达,破坏BTB完整性,增加其通透性,最终影响生精微环境,这可能是GTW致不育的原因之一。而强精片能够降低睾丸内FasL、ABP表达量,提高Occludin蛋白表达,修复BTB功能,可能是中药制剂改善精子功能的机制之一,但在这一过程中中药介导的具体机制尚需进一步研究。
Objective To investigate the effects of QiangJing Tablets on the blood- testis barrier (BTB) function of spermatogenesis microenvironment in infertility rats, and explore its possible mechanism. Mothods Of 70 male SD rats, 20 rats were randomly set as the blank group (BG), and the other 50 rats were induced to be infertility rats by gavage of GTW. After 3 weeks, 10 rats were randomly selected to validate the established model. Forty rats with sterility were randomly recruited and divided into the model group (MG), QiangJing Tablets low-dose group (QJPL), QiangJing Tablets middle-dose group (QJPM), and QiangJing Tablets high-dose group (QJPH). After 4 weeks of drug delivery, their testis were taken and detected. Results Compared with those of BG, the expressions of FasL and ABP in testis of MG were up-regulated significantly (P〈0.01), while the OccludIn was down-regulated remarkably (P〈0.01). Compared with those of MG, the expressions of FasL and ABP in testis of QJPL, QJPM and QJPH were decreased significantly (P〈0.05), especially the expression of FasL in the rats intervened by high doses (P〈0.01). Simultaneously, QiangJing Tablets could improve the expression of Oecludin, compare with MG, the difference were significantly (P〈0.05). Conclusion GTW could increase the expression of FasL and ABP to distract the function of sertoli cell, while inhibit the expression of Occludin. These changes could disrupt the structural integrity of BTB,increase BTB permeability and induce infertility. QiangJing Tablets could down-regulate the expression of FasL and ABP to improve the SC function, meanwhile up-regulate the Occuldin to maintain the structure of tight junction (TJ). More generally, the Chinese herba preparation could repair the structure of BTB destroyed by GTW. But more research was needed to explore the specific mechanism.
