摘要
为考察黄皮果胶的抗疲劳功效,首先采用微波辅助法提取黄皮果胶,通过正交试验获得最优工艺条件为:微波功率600W,提取时间8min,液、料比30∶1,pH值2.0,果胶得率3.61%,总半乳糖醛酸质量分数为78.75%。然后将小鼠分组,分别对其灌胃不同剂量的黄皮果胶,测其体质量,并通过力竭游泳实验,对小鼠的力竭游泳时间,以及血酸、丙二醛、血尿素氮、乳酸脱氢酶、肝/肌糖原等生化指标进行了检测。结果表明:小鼠果胶灌胃2周后,其体质量随果胶剂量的增大而增加,力竭游泳时间随果胶剂量的增大而延长,高剂量时可延长47.95%,具有显著差异。力竭游泳后,小鼠血乳酸、丙二醛和血尿素氮的值均随灌胃果胶量的增加而减小,且高剂量组与对照组有显著差异;而乳酸脱氢酶活力及肝/肌糖原的值则随灌胃果胶量的增加而增大,且中剂量组和高剂量组分别与对照组有显著和极显著差异。黄皮果胶可以有效延缓运动性疲劳,具有良好的抗疲劳功效。
Wampee pectin was extracted firstly using microwave-assisted extraction to investigate the anti-fatigue effect of it.The optimum process conditions optimized by orthogonal test were obtained,of which the microwave powder was 600 W,extraction time was 8min,liquid-to-solid ratio was 30:1mg/L,and pH value of extraction solution was 2.0.The yield was 3.61% and the total galacturonic acid content of pectin was 78.75%.Intragastric administration was performed for different groups of mice using different dose of wampee pectin to determine their body weights,and exhaustive swimming experiment was carried out after two weeks.The exhaustive swimming time and some biochemical indexes such as blood lactic acid,malonaldehyde,blood urea nitrogen,lactate dehydrogenase,hepatic glycogen and muscle glycogen,were subsequently determined.The results showed that the body weights and exhaustive swimming time of mice were increased with the increasing of the pectin.For the mice of high-dose group,the exhaustive swimming time was prolonged 47.95% with significant difference.The values of blood lactic acid,malonaldehyde,and blood urea nitrogen of the mice were all decreased with the increasing of the pectin and there was a significant difference between high-dose group and control group,while the activity of lactate dehydrogenase and the values of hepatic glycogen and muscle glycogen were all increased with the increasing of the pectin,and there were a significant difference between medium-dose group and control group and a highly significant difference between high-dose group and control group.Thus,wampee pectin could delay sports fatigue effectively with good anti-fatigue effect.
