葛根素对极低频电磁辐射下HFSF细胞中MMP-2与COL1A1的影响 被引量:1

Effect of Puerarin on MMP-2 and COL1A1 in HFSF Exposed to ELF-EMFs

摘要 目的:研究葛根素对极低频电磁场(ELF-EMFs)诱导的人胚胎眼巩膜成纤维细胞(HFSFs)中基质金属蛋白酶-2(MMP-2)与I型胶原(COL1Al)的影响。方法:体外培养HFSF细胞,并将其分为对照组、辐射组、葛根素组,通过明胶酶谱法检测MMP-2的活性,Western-Blot检测MMP-2、COL1A1蛋白的表达。结果:暴露于0.2 mT、50Hz的电磁辐射系统24 h后,HFSF细胞(辐照组)MMP-2酶活性较对照组增高20%,MMP-2蛋白表达升高45%,而COL1A1的蛋白表达下降40%,差异有统计学意义(P<0.05);而与辐射组比较,葛根素组HFSF细胞MMP-2酶活性下降33%,MMP-2蛋白表达降低44.1%,COL1A1蛋白升高80%,差异有统计学意义(P<0.05)。结论:极低频电磁辐射可提高HFSF细胞中MMP-2的活性与蛋白表达,抑制COL1Al的合成,葛根素可在一定程度上逆转这一作用。 Objective: To study the effect of puerarin on matrix metalloproteinase-2(MMP-2) and collagen typeⅠ(COL1A1) in human fetal scleral fibroblasts(HFSFs) exposed to extremely low frequency electromagnetic fields(ELF-EMFs). Methods: HFSFs were cultured in vitro and divided into the control group, radiation group and puerarin group. MMP-2 activity were detected by gelatin enzymography. MMP-2 and COL1A1 protein expressions were measured by Western blot. Results: MMP-2 activity and protein expression in radiation group increased 20% and 45%, respectively in comparison with the control group, while COL1A1 protein expression decreased 40%(P〈0.05). MMP-2 activity and protein expression in puerarin group decreased 33% and 44.1%, respectively in comparison with radiation group, while COL1A1 protein expression increased 80%(P〈0.05). Conclusion: ELF-EMFs increased the MMP-2 activity and protein expression in HFSF cells, suppressed the synthesis of COL1A1. Puerarin could reverse the action to some extent.
