摘要
目的探讨红细胞达菲(Duffy)血型和艾滋病病毒(HIV)1型感染的关系。方法将彝族HIV高危人群标本分为实验组(HIV感染者)和对照组(未感染HIV者),应用血清学方法进行Duffy血型表型鉴定,使用聚合酶链式反应-序列特异性引物扩增技术(PCR-SSP)进行基因分型,比较两组Duffy血型表型、基因型的分布及其与该地区已知报道的差异,以及不同Duffy血型表型、基因型的HIV感染率。结果共收集彝族HIV高危人群标本427例,其中HIV感染156例(实验组),未感染166例(对照组)。高危人群Duffy血型分布与同地区同民族差异无统计学意义(P>0.05)。感染者与未感染者Duffy血型差异无统计学意义。表型Fy(a^+b^-)、基因型Fya/Fya、Fya/Fyb/Fy占94.41%(304/322),其中实验组146例,对照组158例;表型Fy(a^+b^+)、基因型Fya/Fyb占4.35%(14/322),其中实验组6例,对照组8例;表型Fy(a^-b^+)、基因型Fyb/Fyb占1.24%(4/322),均为实验组;实验组和对照组均未检出Fy(a^-b^-)表型、Fy/Fy、Fyb/Fyb/Fy基因型。两组Duffy血型表型分布比较差异无统计学意义(P>0.05)。Fya^-与Fya^+高危人群,Fyb^+与Fyb^-高危人群HIV-1感染率比较差异均无统计学意义,基因频率为:Fy^a=0.9239(595/644)、Fy^b=0.0342(22/644)、Fy=0.0419(27/644)。结论血清学、分子生物学方法检测结果一致;流行病学调查方面尚不能认为红细胞Duffy血型与HIV-1感染相关。
Objective To investigate the relationship between erythrocyte Duffy blood group and HIV-1 infec- tion. Methods The samples of HIV high-risk behavior groups among Yi people were divided into an experimental group (infected with HIV) and a control group (not infected with HIV). The two groups were compared for blood phenotype, genotypes and difference of HIV infection in Duffy blood group phenotypes and genotypes by using sero- logical method to identify Duffy blood group phenotyping, and using PCR-SSP technique to assay genotyping. Results There were 427 cases of HIV high-risk behavior groups, with 156 of the experimental group and 166 of the control group. Duffy blood type distribution of the groups had no significantly statistical differences (P〉 0.05) compared with those in the same region and ethnic groups. HIV infected and uninfected Duffy blood group showed no significant difference. Fy (a+b ) phenotype, Fya/Fya and Fya/Fyb/Fy genotype reached 94.41% (304/322), with 146 cases of the experimental group, and 158 cases of the control group. Fy (a+ b+ ) Phenotype, Fya/Fyb genotype were 4.35% (14/322), with 6 cases of the experimental group, and 8 cases of the control group. Fy (a b+ ) Phenotype, Fyb/Fyb genotype were 1.24% (4/322), with all cases of experimental group. There was no Fy (a b ) Phenotype and Fy/Fy, Fyb/Fyb/Fy genotypes. Duffy blood group phenotype distribution of the two groups showed no significant differences, as well as the high-risk of HIV-1 infected group. Fya between Fya+ , Fyb+ between Fyb , the gene frequency was Fya =0. 9239(595/644), Fyb= 0. 0342 (22/644), Fy=0. 0419(27/ 324). Conclusion The results of serological and molecular biology are consistent; and the epidemiological investiga-tions cannot conclude that Dully blood group erythrocytes is associated with HIV-1 infection.
