摘要
河豚毒素主要存在于河豚鱼中,是一种致命的神经毒素。将量子点标记于河豚毒素,并将其作为示踪物,获得高且稳定的荧光信号,建立荧光偏振免疫分析技术检测河豚毒素。所建立的方法检测河豚毒素检测范围为1.36~101.27μg/L,IC50值为11.76μg/L,检测限为0.67μg/L。在该方法的检测范围内标准平均回收率为(99.91±2.60)%,变异系数为2.60%。与竞争性ELISA相比较,本方法河豚毒素测定值与竞争性ELISA测定值呈线性相关,且具有简单、快速等特点,可应用于临床诊断和食品安全检测。
Tetrodotoxin is a fatal neurotoxin which exists in puffer fish.In this study,Quantum dots was linked to TTX as the tracer to achieve high and stable fluorescence signal and a fluorescence polarization immunoassay(FPIA)was developed for the detection of TTX.The developed FPIA for TTX had a detect range from 1.36-101.27μg/L,with an IC50 value of 11.76μg/L and the limit of detection(LOD)value of 0.67μg/L.The recovery test result was(99.91±2.60)%,with the CV value of 2.60%.The developed FPIA had a good correlation with competitive ELISA.The method was simple and fast,which could be applied in clinical diagnoses and food safety detection.
