摘要
华支睾吸虫病(又称肝吸虫病)是一种危害严重的淡水鱼源性人兽共患寄生虫病,吉林省是我国该病危害最为严重的地区之一,为了解吉林省淡水鱼的感染情况,本试验于2014年9月—2015年6月对吉林省扶余市3个乡镇(长春岭镇、得胜镇、更新乡)的松花江流域及沿江支流的野生淡水鱼感染情况进行了形态学观察与PCR鉴定。进行胃蛋白酶消化法的囊蚴形态学观察;PCR引物的设计及特异性鉴定;华支睾吸虫吉林分离株囊蚴和成虫的PCR鉴定。结果显示,3个乡镇分别获得11种淡水鱼,麦穗鱼、泥鳅、黑鱼、老头鱼、鲫鱼、鲶鱼、嘎牙子、白鲢、虾、白鱼、船钉鱼平均每克囊蚴感染量依次为142.16、0.47、0.05、0.38、0.09、0.14、0.32、0.07、0.42、10.23、39.3个,其中麦穗鱼感染量最高。华支睾吸虫吉林分离株18SRNA的PCR扩增片段与中国广西分离株同源性为98%。
Clonorchiasis(also called the liver fluke disease)is a serious hazard of freshwater zoonosis.Jilin province is one of the most serious area in our country,In order to analyse freshwater fish infection situation,wild freshwater fish was collected from three towns(Changchunling,Desheng,Gengxin)of the songhua river basin and along the river tributaries in Fuyu between September2014 and June 2015 for morphological observation and identification of PCR.Pepsin digestion method was used for morphological observation of metacercariae;specific primers was used for PCR identification of metacercariae and adult isolated from Jilin.Average per gram infected metacercariae quantity in Pseudorasbora parva,loach,snakehead,Perccottus glenii,crucian carp,catfish,Pelteobagrus fulvidraco,silver carp,shrimp,white fish,Saurogobio dabryi was 142,0.47,0.05,0.38,0.09,0.14,0.32,0.07,0.42,10.23,39.3,respectively.And Pseudorasbora parva has highest infection quantity per gram.Comparison of PCR amplification fragment of Clonorchis sinensis18 SRNA isolated from Jilin,with gene of Clonorchis sinensis from Guangxi,Zhongguo was 98%homology.
