摘要
目的:研究鞣花酸在Caco-2细胞的摄取特性。方法:采用Caco-2细胞单层模型研究鞣花酸在不同给药时间、药物浓度、p H介质、培养温度、有无抑制剂条件下对细胞摄取的影响。采用高效液相色谱法(HPLC)测定鞣花酸在细胞中的含量。结果:鞣花酸在0.0254~1.016μg范围内呈现良好的线性关系(r=0.9997)。鞣花酸在Caco-2细胞上的摄取与给药时间、药物浓度、p H介质、培养温度、抑制剂密切相关。结论:建立的HPLC检测方法简便、灵敏、准确,鞣花酸在Caco-2细胞上的摄取主要是主动转运,抑制剂不同程度参与鞣花酸的转运过程。
Objective:To study the uptake status of Ellagic acid in Caco-2 cells.Method:Optimization using Caco-2 cell monolayer model research Ellagic acid in different time,concentration,p H,temperature,under the condition of P-gp inhibitors effects on cellular uptake.To establish high performance liquid chromatography(HPLC)determination of the content of Ellagic acid in cells.Result:The linearity of Ellagic acid concentrationcurve was found in the rang of 0.0254~1.016 μg(r=0.9997).Uptake determination of Ellagic acid in Caco-2 Cells absorb a certain amount of time,concentration and p H dependence and Pgp inhibitors can significantly increase the Ellagic acid intake.Conclusion:This method is simple and sensitive HPLC assay can be applied to determine Ellagic acid concentrations in Caco-2 cells.Ellagic acid inhibitors in different level of transfer process.
