静注人免疫球蛋白中砷检测方法的建立

摘要 目的血液制品的包装材料玻璃瓶在存放样品的过程中,有可能有砷溶出,所以需要对制品中的砷进行检测方法的建立。方法用三氯乙酸溶液沉淀静注人免疫球蛋白中的蛋白,分离制品中的砷和蛋白,以氢化物原子吸收法检测血液制品中的微量砷含量。结果 12 mol/L盐酸∶1.2 mol/L碘化钾的体积比为1∶1作为预还原剂时进行预还原,实验效果好,对血液制品中砷的回收率为87.3%-98.7%,仪器定量限为0.914 ng/m L。结论本实验首次建立了血液制品中砷含量的检测方法,该方法简单易行,仪器灵敏度高,实验条件温和。 Objective To establish an arsenic detection method in the blood product packaging materials and glass bottles in the storage process. Method In order to detect the trace arsenic,the arsenic and proteins in blood products were separated using trichloroacetic acid,then the trace arsenic was detected using hydride atomic absorption method. Result The results showed good efficacy using 1. 2 mol / L potassium iodide to achieve prereduction,rendering the 5-valent arsenic reduction to 3-valent under the condition of 12 mol / L HCl. The recovery ratio of arsenic from blood products was 87. 3%-98. 7%,. The limit of quantitation was 0. 914 ng / mL. Conclusion It is the first time to establish a method for detection of arsenic in blood products. The method is easy to operate,sensitive,and has mild experimental conditions.
