血管紧张素转化酶抑制剂通过抗氧化应激对脑梗死大鼠的影响

Therapeutic effects of angiotensin converting enzyme inhibitors on cerebral infarction rats through fighting against oxidative stress

摘要 目的探讨血管紧张素转化酶抑制剂(ACEI)是否可通过抗氧化应激作用发挥治疗脑梗死大鼠的作用。方法实验分假手术组、脑梗死组、福辛普利组、培哚普利组、贝那普利组,后4组建立MCAO模型,造模第3 d开始给药。第1 d、第3 d、第7 d、第14 d、第21 d行Bederson评分;第7 d、第21 d采血检测SOD、MDA、NO;第21 d取脑行TTC染色计算脑梗死体积。结果第14 d,福辛普利组Bederson评分较脑梗死组下降,差异有统计学意义(t=1.930,P=0.044);第21 d,福辛普利组、培哚普利组Bederson评分均低于脑梗死组,差异有统计学意义(t值分别为2.370、2.053,P<0.05)。第7 d,福辛普利组、培哚普利组对比脑梗死组,SOD、NO有所上升,MDA水平下降;第21 d,对比脑梗死组,3个治疗组的SOD、MDA、NO差异均具有统计学意义(P<0.05)。TTC染色后计算发现,培哚普利组梗死体积较脑梗死组缩小。结论 ACEI可能通过抗氧化应激改善大鼠神经功能、缩小梗死面积,从而治疗脑梗死。 Objective Explore the anti-oxidative stress effects of angiotensin converting enzyme inhibitors( ACEI) on cerebral infarction rats. Methods Divide the rats into five groups,including the sham operated group,cerebral infarction group,fosinopril group,perindopril group and benazepril group. The MCAO model was established for the latter 4 groups and medicines were given from the third day. Bederson test was done to score at the 1 d,3 d,7 d,14 d,21 d. On 7 d and 21 d,the levels of serum SOD,MDA and NO were detected. On 21 d,TTC staining of the brain was used to detect the infarct volume.Results The Bederson score of fosinopril group began to decrease compared with the cerebral infarction group on 14 d,and the difference was statistically significant( t = 1. 930,P = 0. 044). On 21 d,the bederson score of the fosinopril group and perindopril group were both lower than the cerebral infarction group,and the differences were statistically significant( t = 2. 370,2. 053,P 0. 05). The serum SOD and NO of the fosinopril and perindopril group were significantly higher than the cerebral infarction group on 7 d while MDA became lower. Compared with the cerebral infarction group,the difference of SOD,MDA and NO between the 3 treatment groups was statistically significant on 21 d( P 0. 05). And the TTC staining told that the infarct volume of the perindopril group was smaller than the cerebral infarction one. Conclusion ACEI may improve the neurological function of the cerebral infarction rats and decrease the infarct volume through anti-oxidative stress to cure the cerebral infarction.
