摘要
采用沸水浸提法提取桑葚多糖,用醇沉法析出粗多糖后利用5%三氯醋酸脱蛋白纯化得精制桑葚多糖。通过正交实验确定最佳提取工艺条件为:料液比1∶6(mg∶mL),浸提时间0.5h,乙醇浓度90%,优化的5%三氯醋酸脱蛋白液料比为875∶1(mL∶g)。在最佳提取工艺条件下进行20倍放大实验,精制桑葚多糖提取率可达2.63%。采用苯酚-硫酸法对桑葚多糖含量进行测定,以葡萄糖溶液为对照品溶液,在490nm处测得吸光度与浓度线性关系良好(R=0.9997);测定溶液在3h内稳定性良好(RSD=1.02%),重复性良好(RSD=1.22%),回收率平均值为99.67%(RSD=0.93%)。该测定方法线性、精密度、准确度均满足测定要求,并且方法简单、成本低,值得广泛应用。
Mulberry polysaccharide was extracted by hot water extraction,crude polysaccharide was obtained by alcohol precipitation method,and protein was further removed by 5% trichloroacetic acid to prepare purified mulberry polysaccharide.Through an orthogonal experiment,the optimal conditions of extraction were as follows:solid-liquid ratio of 1∶6(mg∶mL),extraction time of 0.5h,ethanol concentration of 90%,and 5%trichloroacetic acid with liquid-solid ratio of 875∶1(mL∶g)to deprotein.For 20 times scale-up experiment,the extraction rate of purified polysaccharide could reach 2.63%.Mulberry polysaccharide content was determined by aphenol-sulfuric acid method.Using glucose solution as the reference substance solution,there was a good linear relationship(R=0.9997)between the absorbance at 490 nm and the concentration,the solution had good stability within 3h(RSD=1.02%),It had good repeatability(RSD=1.22%),and the average recovery rate was99.67%(RSD=0.93%).This method meets the requirements of linearity,precision and caauracy,and the method is simple,low-cost and with wide applications.
