祛湿活血方对非酒精性脂肪肝大鼠脂肪组织胰岛素/JNK1通路的影响

Influence of Qushi Huoxue Formula on insuline/JNK1 pathway of adipose tissue in rats with non-alcoholic fatty liver

目的:观察祛湿活血方对非酒精性脂肪肝(NAFLD)大鼠脂肪组织胰岛素/JNK1通路的调节作用。方法:采用高脂饮食喂养12周建立NAFLD大鼠模型,再以祛湿活血方灌胃治疗8周后,对比祛湿活血方组和模型对照组体质量、肝湿重变化,并用Western blotting方法,检测治疗前后脂肪组织胰岛素受体、磷酸化胰岛素受体底物1(p-IRS-1,ser307位点)、c-Jun氨基端激酶1(JNK1)及磷酸化c-Jun氨基端激酶1(p-JNK1)表达的变化,分析祛湿活血方是否影响脂肪组织JNK1表达和活化,是否影响脂肪组织胰岛素信号的转导。结果:祛湿活血方组大鼠体质量及肝湿重较模型对照组明显降低(P<0.05);祛湿活血方组脂肪组织胰岛素受体、1p-IRS-1、JNK1及p-JNK1表达水平较模型对照组无明显变化。结论:尽管祛湿活血方降低了NAFLD大鼠体质量和肝湿重,但是在脂肪组织中并没有改善JNK1信号的变化。

Objective： To observe the influence of Qushi Huoxue Formula on insuline/JNK1 pathway of adipose tissue in rats with non-alcoholic fatty liver disease（NAFLD）. Methods： The NALFD rat model was established by feeding high fat diet for 12 weeks, and then the rats were treated with Qushi Huoxue Formula by gavage administration for 8 weeks. The changes in body weight and liver wet weight of rats in model group and Qushi Huoxue group were compared, the expressions of insulin receptor, phosphorylated insuline receptor substrate 1（p-IRS1, ser307）, JNK1 and p-JNK1 in adipose tissue before and after treatment were detected by Western blotting, and the influence of Qushi Huoxue Formula on the expression and activation of JNK1 and insuline signal transduction of adipose tissue was analyzed. Results： The body weight and liver wet weight of rats in Qushi Huoxue group was obviously lower than that of model group（P0.05）. The difference in expressions of insuline receptor, p-IRS1, JNK1 and p-JNK1 of adipose tissue between Qushi Huoxue group and model group was not significant. Conclusion： Qushi Huoxue Formula could lose the body weight and liver wet weight of NAFLD rats, but it could not improve the changes in JNK1 pathway in adipose tissue.
