摘要
目的:基于四君子汤补脾益气的功效,检测脾虚模型大鼠小肠组织相关基因的表达,探究四君子汤补脾益气作用的生物学机制。方法:将24只大鼠随机分为正常组、脾气虚组、四君子汤组,每组8只,采用PCR array法测定3组间能量代谢相关基因的表达水平。结果:四君子汤促进脾气虚模型大鼠高表达的基因包括嗜铬素A(Chga)、S100钙结合蛋白G(S100g)、双调蛋白(AREG)、糖皮质激素调节蛋白激酶1(SGK1)、生长抑素(SST)、CDKN2B。结论:脾气虚模型能量代谢相关基因的表达与中医脾虚证候密切相关,四君子汤通过促进相关基因的表达,改善脾虚模型大鼠相关证候群。
Objective: Based on the effect of invigorating spleen and replenishing qi of Sijunzi Decoction, to detect the expression of genes in intestinal tissue and explore the biological mechanisms of Sijunzi Decoction. Methods: Twenty-four rats were randomly and averagely divided into normal group, deficiency of spleen qi group, and Sijunzi Decoction group, and the expression levels of energy metabolism related genes of rats in each group were measured by PCR array method. Results: Sijunzi Decoction could promote the high level expression of genes including chromogranin A, S100 calcium binding protein G, amphiregulin, glucocorticoid regulated kinase 1, somatostatin and Cyclin-dependent kinase inhibitor 2B in rats with syndrome of deficiency of spleen qi. Conclusion: The expression of energy metabolism related genes in deficiency of spleen qi model is closely related to the TCM syndrome of spleen deficiency, and Sijunzi Decoction could promote the expression of certain genes to improve the symptoms of rat models with syndrome of deficiency of spleen qi.
