摘要
目的:以长白山区的3、4、5年生西洋参为研究对象,比较西洋参不同部位中4种氧化还原酶的活力。方法:采用p H=7.4的磷酸缓冲溶液从西洋参叶、主根、须根和芦头中提取出氧化还原酶的粗酶液。应用紫外分光光度法分别测定西洋参中过氧化物酶(POD)、过氧化氢酶(CAT)、多酚氧化酶(PPO)和抗坏血酸过氧化物酶(APX)的活力。结果:相同部位的5年生西洋参中氧化还原酶的活力明显高于3、4年生西洋参;4年生西洋参中氧化还原酶的活力略高于3年生西洋参,相同年限的不同部位的西洋参叶中氧化还原酶的活力最大,芦头中酶的活力最小。结论:不同生长年限的西洋参中氧化还原酶的活力差距显著,相同年限中氧化还原酶的活力大小顺序分别为西洋参叶、须根、主根、芦头。
Objective: To compare the activity of oxidation-reduction enzymes in different parts of American ginseng growing in the Changbai mountain with growth years as 3, 4, and 5 years. Methods: The crude enzyme fluid of oxidation-reduction enzymes in leaves, main roots, fibrous roots, and basal part of stem of American ginseng was extracted by using phosphate buffer(p H=7.4). The activity of peroxidase(POD), catalase(CAT), polyphenol oxidase(PPO) and ascorbate peroxidase(APX) was tested by using ultraviolet spectrophotometry. Results: The activity of oxidation-reduction enzymes in American ginseng with 5 growth years was higher than that of American ginseng with 3 or 4 growth years, and activity of oxidation-reduction enzymes in American ginseng with 4 growth years was a litter higher than that of American ginseng with 3 growth years. The activity of oxidation-reduction enzymes in the leaves of American ginseng with the same growth years was the highest, and the activity of oxidation-reduction enzymes in the basal part of stem of American ginseng was the lowest. Conclusion: The difference in the activity of oxidation-reduction enzymes among American ginseng with different growth years is significant, and the activity of oxidation-reduction enzymes in different parts of American ginseng with the same growth years in a descending order as: leaves, fibrous roots, main roots, and basal part of stem.
