强骨胶囊对骨质疏松大鼠OPG/RANKL/RANK系统的影响 被引量:13

Influence of Qianggu Capsule on OPG/RANKL/RANK axial system of osteoporotic rats

摘要 目的:用强骨胶囊对骨质疏松大鼠进行用药干预,观察其对骨质疏松大鼠骨保护素(OPG)、细胞核因子κB受体活化因子(RANK)和细胞核因子κB受体活化因子配体(RANKL)表达的影响,探讨其对骨代谢影响的可能机制。方法:健康雌性SD大鼠,切除双侧卵巢,造成骨质疏松大鼠模型,以低中高不同浓度强骨胶囊(0.054,0.108,0.216g·kg-1·d-1)喂养3个月,并用戊酸雌二醇片0.1mg/kg给药作为阳性对照组,采用荧光定量技术检测破骨细胞OPG、RANK和RANKL的表达。结果:阳性对照组及各治疗组均可使OPG的表达量增加,RANK和RANKL的表达量减少(P<0.05),且随着骨碎补总黄酮浓度的增加,OPG表达呈上升趋势,RANK和RANKL表达均呈下降趋势。强骨胶囊上调表达OPG m RNA,同时下调表达RANKL m RNA和RANK m RNA,使RANKL/OPG的比值下降,这种调节作用在高剂量(0.216g·kg-1·d-1)作用时最明显。结论:强骨胶囊可影响大鼠OPG、RANKL和RANKL m RNA表达,调节RANKL/OPG比值,从而影响骨代谢。 Objective: To treat osteoporosis rats with Qianggu Capsule, and observe the influence of Qianggu Capsule on expression of osteoprotegerin(OPG), receptor activator of NF-κB(RANK) and receptor activator of NF-κB ligand(RANKL) of osteoporosis rats. Methods: Female healthy SD rats were used to establish the osteoporosis models by resecting bilateral ovaries. The model rats were fed with Qianggu Capsule with different concentration(0.054, 0.108, 0.216g·kg-1·d-1) for 3 months, and the rat models fed with USP(0.1mg/kg) were served as the control group. The expression of OPG, RANK and RANKL was detected by using RT-PCR technology. Results: USP and osteopractic total flavonoids with different concentrations could increase the expression of OPG and decrease the expression of RANK and RANKL(P0.05). As the concentration of osteopractic total flavonoids increased, the expression of OPG was on the increase, and the expression of RANK and RANKL was on the decrease. Qianggu Capsule could up-regulate the m RNA expression of OPG, down-regulate the m RNA expression of RANK and RANKL, reduce the ratio of RANKL/OPG, and the regulation effect of high dosage(0.216g·kg-1·d-1) was the most obvious. Conclusion: Qianggu Capsule could affect the m RNA expression of OPG/RANKL/RANK, regulate the ratio of RANKL and OPG, and influence bone metabolism.
