摘要
目的:用细胞学方法评价左金丸、反左金丸、茱萸丸、甘露散、交泰丸等中药复方的寒热药性。方法:用噻唑蓝(MTT)比色法考察5种中药复方对He La细胞和SGC-7901细胞体外增殖的影响,倒置显微镜观察其对细胞形态特征的影响。结果:反左金丸、茱萸丸在所选质量浓度5-800μg/m L范围内对He La细胞和SGC-7901细胞的生长增殖表现出促进作用,表现为热性;左金丸、甘露散、交泰丸在所选质量浓度5-800μg/m L范围内对He La细胞和SGC-7901细胞的生长增殖表现出抑制作用,随着浓度的增大,抑制率逐渐增大,表现为寒性。形态学观察表明,左金丸、甘露散与交泰丸能使细胞密度减小,固缩变圆;反左金丸、茱萸丸可使细胞密度增加,结构致密,生长旺盛。台盼蓝染色表明,5种中药复方对这两种细胞没有细胞毒作用。结论:5种中药复方的寒热药性细胞学方法评价结果与文献记载一致,说明细胞学方法能够用于中药复方寒热药性的评价,为研究中药复方寒热药性提供了实验依据和方法学参考。
Objective: To evaluate the cold-heat nature of Zuojin pill, Fanzuojin pill, Zhuyu pill, Ganlu powder and Jiaotai pill by using cytological method. Methods: MTT assay was used to observe the effect of the five TCM compound prescriptions on the proliferation of He La cells and SGC-7901 cells in vitro. Morphological changes were observed by using inverted microscope. Results: Fanzuojin pill and Zhuyu pill could promote the growth and proliferation of He La cells and SGC-7901 cells within the range of concentrations as 5-800μg/m L, which demonstrated that the drug property of Fanzuojin pill and Zhuyu pill was heat. On the other hand, Zuojin pill, Ganlu powder, and Jiaotai pill could inhibit the growth and proliferation of those cells within the range of concentrations as 5-800μg/m L, and with the increasing of concentration, the inhibition rate also increased gradually, which demonstrated that the drug property of the above 3 formulas was cold. Morphological observation demonstrated that Zuojin pill, Ganlu powder and Jiaotai pill could reduce the cell density, caused pycnosis in cells, and make cell turn round, while Fanzuojin pill and Zhuyu pill could increase the ell density, enhance the cellular structure and promote cell's growth. The results of trypan blue staining demonstrated that 5 compound prescriptions had no cytotoxic effect on the 2 kinds of cells. Conclusion: The coldheat nature of the 5 compound prescription demonstrated in this study was in accordance with literatures, which showed that the cytological method could be applied in evaluating the cold-heat nature of TCM compound prescriptions, and provide experimental bases and methodology reference for the studies about cold-heat nature of TCM compound prescriptions.
